其他摘要Chitinases and B-1, 3-glucanases were purified from the primary and secondary corm of Gastrodia elata by precipitation with ammonium sulfate, chromato-graphies on activated charcoal, sephadex G-50, DEAE-sephadex and hydroxyapatite column. Their yields in primary corm, 0.4 and 0.2 mg/100g FW respectively, were ca. 3 times higher than those in secondary corm. The chitinase and B-1, 3-glucanase from primary corm, which is infected by Armillarilla mellea and supplies with nutrition for primary corm and secondary corm, were compared with those from secondary corm, which cannot be infected by A. mellea, in physical property, enzymic activity and antifungal activity. On SDS-PAGE, the chitinases and B-1, 3-glucanases from primary corm, mol wt 31.5 and 94.0 KD respectively, and secondary corm, mol wt 34.5 and 91.0 kD, all showed a single band. The specific activities of the hydrolases from primary corm were ca. 30 times higher than those from secondary corm, but their antifungal activities were ca. 7 times higher than the secondary hydrolases's. The contribution of the chitinase and B-1, 3-glucanase in primary corm to G. elata's defense against pathogen is only one seventith of GAFP's. This suggests that the hydrolases are not effective antifungal agent but GAFP. The hydrolases in primary corm are thought to partially digest the growth hyphae tip of A. mellea.
