摘要 | In the present study, a series of copper (II) complexes containing 1,8-naphthalimide moiety (complexes 1-3) were synthesized and structurally characterized by Infrared spectroscopy, elemental analysis, high-resolution mass spectrometry, and single crystal X-ray diffraction. The results of single crystal diffraction show that the copper ions in three complexes are five-coordinated with three N atoms of Di-(2-pyridylmethyl)amine (DPA) and two O atoms of acetylacetonate, forming a distorted trigonal bipyramidal geometry. The in vitro cytotoxicity of three copper(II) complexes was evaluated by MTS assay against five cancer cells, the result displayed that the anticancer activity of the compounds in this series was regulated by the aliphatic chain length of ligands with IC50 ranging from 2.534 mu M to 19.18 mu M, their anticancer activity became more potent as the aliphatic chain length of ligands increased. Complex 3 displayed the most potent antiproliferative activity among the three complexes, particularly for MDA-MB-231, being nearly 10 times higher than that of cisplatin. Complex 3 exhibited a superior capacity to trigger apoptosis in MDA-MB-231 cells compared to cisplatin, and caused the dose-dependent cell cycle arrest. In addition, the DNA interaction of ligand 3 and complex 3 were investigated by UV absorption spectroscopy, fluorescence spectroscopy and viscosity measurements, the results showed that complex 3 had a high affinity for binding to DNA in an intercalating mode, but ligand 3 interacted with DNA through an electrostatic interaction. The molecular docking studies of complex 3 with duplex DNA further suggested that the complex 3 intercalated into base pairs of DNA. |
