摘要Snails are not only a delicacy on the table, but also have high medicinal value. Snail polysaccharide is one of the most important active ingredients of snails, including neutral sugars and glycosaminoglycans. The glycosaminoglycans of mollusks are rich in sulfate groups, which have a wide range of pharmacological activities such as anti-thrombosis, anti-virus, anti-inflammatory, and immunity enhancement, especially in wound healing and tissue repair. In this paper, the polysaccharides in the gastropod tissue of Helix lucorumwere separated and purified, two glycosaminoglycans and one neutral sugar were prepared, carry out physical and chemical properties analysis and chemical structure research to evaluate the immune regulation of snail polysaccharides and the pharmacological activity of promoting wound healing, laying a foundation for the comprehensive utilization of snail polysaccharides and snail resources. Enzymatic hydrolysis-alkaline hydrolysis was used to extract polysaccharides from the soft tissues of the artificially reared Helix lucorum gastropod, and purified by deproteinization, salting out and alcohol precipitation, quaternary ammonium salt precipitation, and ion exchange to obtain two snail glycosamines with different structures. The yields of glycans HL-1 and HL-2 were 0.2% and 0.6%, respectively. Analysis of physicochemical properties such as monosaccharide composition, optical rotation, molar ratio of sulfuric acid and carboxylic acid of HL-1 and HL-2, as well as UV, IR and NMR spectrum analysis results show that HL-1 is a glycosaminoglycan (Mw 6.20×105 Da) composed of acetylglucosamine (GlcNAc), acetylgalactosamine (GalNAc), L-iduronic acid (IdoA) and galactose (Gal)., HL-2 (Mw1.34×105Da) is composed of acetylglucosamine, L-iduronic acid and galactose.The physical and chemical properties such as the molecular weight and sulfate group content of the two are different, but the structure of the glycosaminoglycan is extremely complex, and the exact chemical structure needs to be further studied. Purification also resulted in a neutral sugar HL-3. The composition analysis of the monosaccharide showed that it contained only glucose; technical methods such as methylation analysis, 1D and 2D NMR analyzed the chemical structure of the polysaccharide, which was expressed as α(1→4) glycosidic bond is the main chain, and the α-D-glucan is substituted with the α(1→6) glycosidic bond glucose (Glc) side chain. The mouse macrophage cell line RAW 264.7 was used to evaluate the survival rate and phagocytic ability of snail polysaccharide SP on macrophages. The results showed that the sugar promotes the proliferation of macrophages, significantly enhances the phagocytic function of phagocytes, and can promote macrophages NO release and significantly stimulate macrophages to produce TNF-α.
