摘要 | Euphorbia peplus, a plant of genus Euphorbia (family Euphorbiaceae). It is a temperate annual weed that grows up to 40 cm tall. It was originally native to Europe and North Africa, but is now almost cosmopolitan. The plant has always been used as a folk medicinal herb, often used to treat skin lesions, inflammatory diseases, asthma, diabetes and cancer. In order to further enrich the chemical composition diversity of Euphorbia peplus and to explore its biological activity in depth, this thesis investigates the chemical composition and its biological activity of Euphorbia peplus harvested from around Kunming. The thesis consists of four chapters. The first chapter is a study on chemical composition of Euphorbia peplus. 47 compounds were isolated and identified, including 45 diterpenoids, 14 of which were new compounds. Diterpenes consist of 27 jatrophane diterpenes, 12 ingenane diterpenes, 1 abietane diterpene, 2 paraliane diterpenes, 1 lathyrane diterpenes, 2 pepluane diterpenes. To obtained compounds, a variety of separation methods were used, such as MCI gel, silica gel, Sephadex LH-20, Styrogel, reverse phase C-18 silica gel and high performance liquid chromatography (HPLC). Their structure and relative configuration were determined by spectroscopic analysis, physicochemical characteristics, and X-ray diffraction analysis. The second chapter detailed the bioactivities of the compounds of Euphorbia peplus. The activity of the autophagy flow of HM mCherry-GFP-LC3 cells was evaluated, and the activity of the new compound 14 to enhance lysosomal biogenesis was evaluated by LysoTracker Red staining and western blot analysis. It was found that compound 14 could promote lysosomal biogenesis. Western blot analysis was used to evaluate the anti-Aβ activity of ingenane-type compounds and to explore the structure-activity relationship. The third chapter detailed the chemical composition of Daphniphyllum angustifolium Hutch.. Using various separation methods, modern spectroscopy techniques and other methods, two Daphniphyllum alkaloids were isolated and identified from the stem of Daphniphyllum angustifolium Hutch.. The fourth chapter detailed a review of the progress of research on the chemical composition and related pharmacological activities of Euphorbia peplus, including the main chemical composition and related biological activities. Biological activity studies have shown that Euphorbia peplus has a certain anti-inflammatory effect, P-gp inhibition, irritation and other activities. |
