摘要 | Lepidium meyenii(maca)comes from Peru, South America, which was introduced into China for cultivation in 2002, then maca industry of Lijiang, Yunan has achieved rapid growth. In recent years, a series of special hydantoins and thiohydantoins have been isolated from maca for the first time, and the thiourea and urea fragments of its structure are an important class of pharmacodynamic fragments. Scleropyrum wallichianum is a native plant distributed in Xishuangbanna, Yunnan, which has a long history of eating. The analysis of kernel oil of S. wallichianum found that the oil has high degree of unsaturation, and contains lots of characteristic acetylenic acid components. In order to further clarify their characteristic components, the characteristic chemical compounds of maca and S. wallichianum were systematically studied through HPLC-UV, 1D NMR, UPLC-MS/MS. Detailed research contents are as follows: Lepidium meyenii (Brassicaceae), a series of hydantoins and thiohydantoins have been isolated from maca in recent years. Researches show that thiourea and urea fragments are important pharmacodynamic fragments in the study of antitumor drugs. Therefore, our research purpose is to excavate deeply (thio)hydantoin compounds from maca. In the present study, HPLC-UV associated with 1D NMR were used to track (thio)hydantoins from maca, and 39 compounds were separated and identified, of which 14 were new compounds, including 3 pairs of rare disulfide bond-bearing macathiohydantoin dimers, 8 pyrrole alkaloids (including 2 dimers), 2 macamides, and 1 thiohydantoin. Bioactive assay studied showed that disulfide bond-bearing macathiohydantoin dimers had significant protective effect on PC12 cell damaged by corticosterone, with the highest cell survival rate 72.64%, compared with 88.49% of the positive control Desipramine. Compound 4 had a selective inhibitory effect on the liver cancer cell line SMMC-7721, with an IC50 of 16.87±0.49 μM, which is equivalent to the positive control DDP (IC50 11.30±0.50 μM). Scleropyrum wallichianum (Santalaceae) is a small evergreen tree, and its kernel is a traditional edible part of ethnic minorities in Yunnan. The analysis of seed oil of S. wallichianum found that a number of acetylenic acids were present in seed oil. Alkyne compounds are an important class of metabolites in nature, and have various biological activities. However, there are few studies on the acetylenic acid in S. wallichianum. Therefore, 1D NMR and UPLC-MS/MS were used to track the acetylenic acid in the ethyl esterified seed oil, and 14 acetylenic acid were isolated, including five new acetyleneic acid featuring an acetyenic ketone fragment. Their structures were confirmed by 1D NMR, TOCSY spectra, and UPLC-MS/MS analysis. Then the characteristic secondary fragment ions of compounds 1-10 were summarized, which provide the foundation for further separation and analysis of acetylenic acids. |
