摘要 | Corybas taliensis Tang & F. T. Wang is a terrestrial orchid from Ochidaceae with extremely high ornamental value. The hole plant is only 5 – 6 cm high with one exceptionally beautiful fascinating colourful flower and one heart-shaped leaf. C. taliensis always grows among mosses, which is sensitive to the environment changes, and thus the geographical distribution of this species is extremely limited. The species is facing a high risk of extinction with the distribution range reduction under nowadays global warming and human disturbance. Thus, C. taliensis has been categorized as Endangered (EN) in Red List of Biodiversity in China (2013), and also in Threatened Species List of China’s Higher Plants (2017) published with the Chinese journal Biodiversity Science. The species was also listed as potential target species in China’s national program of “Survey and Germplasm Conservation of Plant Species with Extremely Small Populations (PSESP) in Southwest China (2017FY100100)”. To provide research basis for conserving C. taliensis scientifically, the systematical investigations on the geographical distribution and populations, and assessment of the threatened status have been carried out. Then the characteristics of complete chloroplast genome, genetic diversity and genetic structure based on restriction site-associated DNA sequencing (RADseq), pollination biology , and the in vitro seed germination of C. taliensis have also been conducted in this study. The major results are as follows: 1.Population biology, habitat characteristics and threatened status assessment Investigation of eight C. taliensis populations and one C. fanjingshanensis population (which is morphologically similar to C. taliensis) showed that they always grow among mosses under the Rhododendron species, or wet leaf litter and rotten wood sometimes at an altitude between 2200 m and 3200 m. The total area of occupancy of these populations is some 0.04 km2. There are five populations of C. taliensis within in nature reserve and they are almost free from human disturbance, while the other three outside the nature reserve are under moderate or high levels of disturbance caused by grazing, road construction, wind farm and mosses collecting. There are four populations with more than 2000 individuals, but the other four populations with less than 600 individuals are facing a risk of extinction. As the number of populations and mature individuals of C. taliensis are not very small, but the area of occupancy is very limited. And therefore, the threatened status of C. taliensis should be reassessed as Vulnerable (VU) according to IUCN Red List Categories and Criteria (2012). 2.Complete chloroplast genome characteristics, genetic diversity and structure Complete chloroplast genome of C. taliensis was assembled using Illumina pair-end sequencing data from leaf DNA, then phylogenetic tree based on 13 chloroplast genomes of Orchidoideae was built.
