牛肝菌科绒盖牛肝菌亚科的分子系统发育研究; Molecular phylogeny of Xerocomoideae in Boletaceae

摘要The subfamily Xerocomoideae belongs to the fungal family Boletaceae and is of significant ecological importance, forming ectomycorrhizal symbiosis with plants of more than 10 families such as Fagaceae, Pinaceae and Leguminosae. This subfamily has a high species diversity and the complexed morphological characters, which make it be a problematic taxonomic group. By means of the molecular phylogenetic analyses, the previous studies outcame some important achievements. However, due to the limited DNA fragments causing the insufficient phylogenetic informative sites, and the incomplete sampling, the specie diversity and the phylogenetic relationships among most of the genera and the major clades were not very clear. Based on comprehensive sampling and specimen loan from domestic and abroad herbaria, this study screened a batch of single-copy nuclear genes, and implemented the next-generation sequencing technique for molecular phylogeny. We aimed to reconstruct the molecular phylogenetic framework of Xerocomoideae and uncover the evolution of the basidiospore ornamentations in the subfamily. Firstly, 51 single-copy orthologous genes were screened out from 96 ones, and 101 species, 181 samples were sequenzed. In totoal, 7762 sequences were newly generated. By using the methods of Maximum Likelihood and Bayesian Inference, the clear molecular phylogenetic framework of Xerocomoideae were reconstructed, in which the phylogenetic relationships among most of genera and major clades were well resolved. In general, the subfamily Xerocomoideae can be subdivided into two major clades and 12 generic clades. One of the two major clades consisted of Aureoboletus, Hemileccinum, Hemioporus and Hongoiboletus gen. nov., and its diagnosis character is that most of the species in this major clade lack of discoloration when hurt. The other major clade comprised Phylloporus, Hourangia, Xerocomus, Boletellus and other 4 potential new generic clades and its distinguishable character is that almost all species in this major clade can change blue when hurt. In addition, this study verified that all the known genera, namely Aureoboletus, Hemileccinum,Heimioporus, Phylloporus, Hourantia, Xerocomus, and Boletellus, are monophyletic. Secondly, by comprehensive sampling, this study for the first time inferred the African origin of the main clade including genera with bacillate basidiospore ornamentation, namely Phylloporus, Hourangia, Xerocomus and other 4 potential new generic lineages. After the origination, the species in this main clade probably firstly diversified in Africa because its basal four clades were from Africa. When the species spread into East Asia, especially China, this main clade probably undergone the rapidly radiated speciation because the extremely high diversity of this main clade in China. Thirdly, we found species in the subfamily Xerocomoideae evolved at least four main types of basidiospore ornamentations: pinholed (Hemileccinum), bacillate (Phylloporus,
