摘要 | Thrips-transmitted Tomato spotted wilt virus (TSWV),the type member of the genus Tospovirus of the family Bunyaviridae, It causes seriously damage worldwide to many important cash crops such as tobacco, pepper and Tomato..Therefore, it is listed as one of the top ten plant viruses in the world. Due to development of insecticide resistance,pesticide residue, varieties resistance breakout or lack, efficitive method to control the virus disease is lack. Compared with chemical control, botanical antiviral agents have the advantages of safety and environmental protection, low toxicity or non-toxicity, easy degradation and resistance. Therefore, it is very important to search for plant materials with potential anti-tswv activity and elucidate the active substance basis to control TSWV This thesis investigated the chemical constitutes and biology activities of C21 steroidal glycosides from the roots of Cynanchum paniculatum (Bunge) Ktag. Using a variety of separation methods, thirty-one compounds, including nine new ones, were isolated, and all of compounds belonged to C21 steroidal glycosides. Their structures were elucidated by spectroscopic analyses, including extensive 2D NMR and acid hydrolysis. The anti- Tomato spotted wilt virus (TSWV) activity of part of those C21 steroidal glycosides were screened by the conventional method of lesion counting, and certain compounds of the above have potent protective and curative activities. In addition, we also tested the cytotoxic activities of some of the C21 steroidal glycosides. The research progress of C21 steroidal glycosides and anti- Tomato spotted wilt virus (TSWV) was also reviewed. Chapter 1 reviewed the C21 steroidal glycosides, including C21 steroidal glycosid- es’ sources, structure and their biological activities. We also summarized the distribution, structure, damage and prevention of TSWV and antiviral agent from natural products. Chapter 2 reported the results of the phytochemistry study of Cynanchum panic- ulatum (Bunge) Ktag.Thirty-one compounds were isolated from the roots of Cynanc- hum paniculatum (Bunge) Ktag, including nine new ones ,and all of compounds belonged to C21 steroidal glycosides. Chapter 3 reported biology activities of C21 steroidal glycosides isolated from Cynanchum paniculatum (Bunge) Ktag. Antiviral activities of the C21 steroidal glycosides showed that some compounds had significant activities against TSWV. In addition, ELISA a showed that compounds 15, 16, 19, 24 and 30 had significant activities against TSWV. Other compounds also have some preventive activity. In this part, the anti-TMV activity of 49 compounds isolated from Wedelia trilobata (L.) Hitchc. (34-59,70-82) and Manglietiastrum sinicum (Y. W. Law) (60-69) were also investigated. In addition, some compounds were screened for cytotoxic activity in vitro, and compound 21 showed weak cytotoxic activity in vitro against leukemia cells, liver cancer cells, breast cancer cells and colon cancer cells.
