英文摘要: The thesis consists of four chapters altogether. The first chapter shows the isolation and identification of all kinds of compounds from Limonia crenulata Roxb. The second chapter presents the structures of coumarins from a famous Chinese traditional medicine-//e/-ac/ewm rapula Franch. Bioactivity toward platelet aggregation induced by AA, ADP, or PAF was also described. The third chapter clarifies the diterpenoids isolated from three Isodon species, Isodon eriocalyx (Dun) Hara var. laxiflora C. Y. Wu et H. W. Li, Isodon xerophilus (C. Y. Wu et H. W. Li) H. Hara, and Isodon glutinosa (C. Y. Wu et H. W. Li). In this chapter, the antitumor activity of dierpenoids from Isodon eriocalyx var. laxiflora was reported and the structure-function relationship of these compounds was briefly discussed. From the five medicinal plants above, 113 compounds were purified and identified, which concern diterpenoids, alkaloids, coumarins, flavanones, tetranortriterpenoids, triterpenoid, steroids, lignans, aromatic compounds and ceramides. Among them, 22 compounds were proved to be new ones, including one alkaloid, three coumarins, three abietane diterpenoids, one isopimarane diterpenoid, two novel skeletal e"/-kauranoids, and twelve eK^-kauanoids. In the last chapter, new naturally occurring coumarins isolated from 1997-Nov. 2001 were summarized. Chapter 1. Study on Chemical Constituents of Limonia crenulata Limonia crenulata was widely cultivated in Myanma, as an industrial crop. From the late 50's of 20th century, scientists in southeastern Asian countries especially in India, Pakistan and Sri Lanka, have extensively investigated phytochemicals of this plant. In order to search active constituents from Limonia crenulata, we studied phytochemically on the stems of this plant, which was purchased from Dehong prefecture of Yunnan Province. As a result, 23 compounds were isolated and identified, which included a simple indole alkaloid, a quinoline alkaloid, two amide alkaloids, four coumarins (including a dimer), two flavanoids, three tetranortriterpenoids, one tripenoid, five steroids, two lignans and two aromatic compounds. Compound 1 was a new simple indole alkaloid. Compounds 4-8, 10, 12, 14, 17-19 were obtained from this plant for the first time. Their structures were determined on the basis of spectral evidence. Compounds from Limonia crenulata are listed as follows, and their structures are illustrated in Figure 1. Study on Chemical and Bioactive Constituents of Heracleum rapula Heracleum rapula is a Chinese traditional medicine. Many studies on chemical constituents of this plant have been reported in recent years. It has been shown that liposoluble fraction of this plant extract is rich in furanocoumarins. Investigation on polar fraction has been rarely reported. From the fresh roots of Heracleum rapula collected in Dali prefecture of Yunnan province, 23 compounds including 20 coumarins were isolated and characterized. Five coumarin glucosides, including two new ones, were obtained from the water-soluble fraction. The other coumarins isolated from the ethyl acetate-fraction included ten furanocoumarins, a simple coumarin and three types of bicoumarins. Their structures were elucidated by means of spectroscopic analysis. Compound 1, a new coumarin glucoside, in which the glucose was directly linked to the coumarin nucleus with no further in between, was quite different from the other four ones, which was found to be rare in naturally occurring coumarins. Compound 2 contains two sugar units. It was noteworthy that, compound 3 and its aglycone (13), were found to be rich in aqueous and EtOAc parts respectively. However, a considerable amounts of 4 and 5 have been isolated from the aqueous fraction, neither of their corresponding aglycones were obtained from the EtOAc fraction, even in trace amounts. This might be one of the important reasons that resulted in the different bioactivities of its two parts (EtOAc and H2O). In our study, a large amount of ceramides was also obtained. The structures were determined on the basis of spectral evidence and chemical reactions. The biological activity assays of all compounds toward platelet aggregation induced by AA, ADP, or PAF were also carried out. Compound 11 exhibited significant inhibitory activity on AA-induced platelet aggregation in a concentration-dependent manner in vitro, with IC50 value of 23.6±4.5 p.M which was better than that of aspirin (IC50: 35.2±8.6 uM).
