题名: 苍白秤钩风、秤钩风的化学成分与抗HBV活性研究

中文摘要: 本论文前两章报道了2种药用植物苍白秤钩风 (Diploclisia glaucescens) 和秤钩风 (Diploclisia affinis) 的化学成分的研究结果,第三章报导了从苍白秤钩风和秤钩风中分离得到的部分化合物进行抗HBV活性研究的结果,第四章综述了2000年以来国内外对具有抗HBV活性的天然化合物的研究概况。 利用各种色谱技术,从这两种植物中共分离得到27 个化合物。利用各种波谱数据分析,鉴定了化合物结构。抗HBV活性研究发现,其中6个化合物具有一定的抗HBV活性。 第一章 苍白秤钩风的化学成分研究 苍白秤钩风Diploclisia glaucescens (Blume) Diels为防己科 (Menispermaceae) 秤钩风属植物的木质藤本。我们对采自西双版纳的苍白秤钩风藤茎的化学成分进行了研究,采用多种柱层析方法从中分离到了17个化合物。经波谱学方法鉴定为:2, 3, 10, 11, 12 - pentamethoxy - 8 - oxoberbine (1)、gusanlung C (2)、(+) - N - carboxamidostepharine (3)、 promucosine (4)、N - propylstepharine (5)、orientaline (6)、 3′ - hydroxyl - N - methylcoclaurine (7)、coclaurine (8)、longifolonine (9)、 3 - methoxyacridan (10)、abutasterone (11)、 paristerone 20, 22 - monoacetonide (12)、polypodine B 20, 22 - acetonide (13)、ponasterone A (14)、pinnatasterone (15)、 旌节花甾酮A (stachysterone A, 16) 及β-谷甾醇 (β - sitosterol, 17)。其中化合物1, 3, 4, 5, 7, 9, 10, 15, 16为首次从该植物中分离。 第二章 秤钩风的化学成分研究 秤钩风Diploclisia affinis (Oliv.) Diels为防己科 (Menispermaceae) 秤钩风属植物的木质藤本。国内外尚无对该种植物的化学成分的研究报道,我们对采自西双版纳的秤钩风藤茎进行了化学成分研究,从中分离到了10个化合物,包括异喹啉生物碱和甾酮类化合物。经波谱学方法鉴定为:瑞枯灵 (reticuline, 18)、N - 降菏叶碱 (N - asimilobine, 19)、acutumine (20)、2, 3 - 二羟基 - 9, 10 - 二甲氧基四氢原小檗碱 (dihydroxyprotoberberine, 21)、千金藤啶碱 (stepholidine,22)、2β, 3β, 20β, 22α, 25 - 五羟基 - 8, 14 - 二烯 - 胆甾 - 6 - 酮 (2β, 3β, 20β, 22α, 25 - pentahydroxy - cholesta - 8, 14 - dien - 6 - one,23)、20 - hydroxyecdysone (24)、旌节花甾酮B (stachysterone B, 25)、calonysterone (26)、旌节花甾酮C (stachysterone C, 27)。所有化合物均为从该植物中首次分离得到。 第三章 苍白秤钩风和秤钩风的抗HBV活性研究 对两种植物的部分化合物进行抗HBV活性研究,研究发现:苍白秤钩风中化合物ponasterone A对HBsAg和HBeAg均显示较好的抑制活性,其对HBsAg对HBeAg的IC50分别为0.37 mM和0.56 mM; SI分别为>8.24和>5.38。化合物gusanlung C、 paristerone 20, 22 - monoacetonide及polypodine B 20, 22 - acetonide仅对HBsAg有一定的抑制作用,IC50分别为1.53, 0.36, 0.24 mM; SI分别为1.58, 1.16, 2.98。秤钩风中化合物2β, 3β, 20β, 22α, 25 - pentahydroxy - cholesta - 8, 14 - dien - 6 - one对HepG2.2.15细胞分泌HBsAg的抑制效果较好 (IC50 = 0.24 mM, SI > 12.0), 化合物瑞枯灵 (reticuline) 对HepG2.2.15细胞分泌HBsAg有一定的抑制效果 (IC50 = 0.58 mM, SI > 1.13)。 第四章 抗HBV 活性天然化合物研究进展 本章综述了2000年以来国内外已经发表报道的包括木脂素和多酚类、黄酮类、生物碱类、香豆素和醌类、萜类及多糖类物质的总共95个化合物抗HBV活性研究概况。
英文摘要: The thesis is composed of four chapters, in which the chemical constituents of two medical plants, Diploclisia glaucescens and Diploclisia affinis had been investigated. The third chapter focused on the bioassay of anti-HBV compounds from the two plants. In the last chapter, the progress of the active anti-HBV compounds from natural products was summarized based on literatures from 2000 to 2008. 27 compounds were isolated from the above two species and elucidated based on extensive spectral analyses (1D, 2D, MS, IR). Bioassay on the partly compounds showed that six compounds presented anti-hepatitis B virus activities. Chapter 1 The Chemical Constituents from Diploclisia glaucescens Diploclisia glaucescens is a folk herb used for the treatment of rheumatism, urethra inflammation and snakebite. Seventeen compounds were isolated from the species, whose structures have been elucidated as 2, 3, 10, 11, 12 - pentamethoxy - 8 - oxoberbine (1)、gusanlung C (2)、(+) - N - carboxamidostepharine (3)、 promucosine (4)、N - propylstepharine (5)、orientaline (6)、 3′ - hydroxyl - N - methylcoclaurine (7)、coclaurine (8)、longifolonine (9)、3 - methoxyacridan (10)、abutasterone (11)、 paristerone 20, 22 - monoacetonide (12)、polypodine B 20, 22 - acetonide (13)、ponasterone A (14)、pinnatasterone (15)、stachysterone A (16)、 β - sitosterol (17). Chapter 2 The Chemical Constituents from Diploclisia affinis Diploclisia affinis belongs to the Menispermaceae family. Chemical investigation on D. affinis led to the isolation of ten compounds for the first time. The structures of compounds isolated were elucidated as reticuline (18)、N - asimilobine (19)、acutumine (20)、dihydroxyprotoberberine (21)、stepholidine (22)、2β, 3β, 20β, 22α, 25 - pentahydroxy - cholesta - 8, 14 - dien - 6 - one (23)、20 - hydroxyecdysone (24)、stachysterone B (25)、calonysterone (26)、stachysterone C (27). Chapter 3 Acitive Anti-HBV Constituents from Diploclisia glaucescens and D. affinis Anti-HBV bioassay on selected compounds from the two plants, Diploclisia glaucescens and Diploclisia affinis was investigated using HepG2.2.15 cell line. Six active compounds were revealed. Ponasterone A from Diploclisia glaucescens, showed good inhibitions on HBsAg and HBeAg secretions (IC50 = 0.37 mM, 0.56mM; SI = >8.24, >5.38, respectively). Gusanlung C、paristerone 20, 22 - monoacetonide and polypodine B 20, 22 - acetonide from Diploclisia glaucescens showed good inhibitions on HBsAg secretion (IC50 = 1.53, 0.36, 0.24 mM, and SI = 1.58, 1.16, 2.98, respectively). 2β, 3β, 20β, 22α, 25 - pentahydroxy - cholesta - 8, 14 - dien - 6 - one from Diploclisia affinis showed good inhibition on HBsAg secretion (IC50 = 0.24 mM, SI > 12.0). Reticuline showed inhibition on HBsAg secretion (IC50 = 0.58 mM, SI >1.13). Chapter 4 Review on Active Anti-HBV Compounds from Natural Products 95 active anti-HBV compounds from natural products were summarized based on literatures from 2000 to 2008.
