中文摘要: 内生菌是天然产物的重要来源,但是目前对其研究相对较少。本论文以三株植物内生链霉菌及珍贵橙色束放线菌突变株HGF052为研究对象,试图从其发酵产物中寻找结构新颖、具有生物活性的天然产物。 论文共由五章组成,其中第一章主要介绍植物内生菌的分离、筛选与鉴定。从滑桃树内生菌中筛选出三株次生代谢产物相对较为丰富的菌株W5、M4、4A8y;经16S rDNA序列鉴定,W5、M4、4A8y均属于链霉菌属(Streptomyces)。 第二章主要研究滑桃树内生链霉菌W5、M4的次生代谢成分,从中共分离得到10个化合物,分别是:cis-2-(1′,8′-dihydroxy-8-methylnonyl)-3-hydroxyl- methylbutanolide (1)、5,10,11-cadinanetriol (2)、dimeric dinactin (3)、(+/−)-nonactic acid (4/5)、(R)-2-((2R,5S)-tetrahydro-5-(2-oxopropyl)furan-2-yl)propanoic acid(6)、2-(5-(2-acetoxypropyl)-tetrahydrofuran-2-yl)propanoic acid (7/8)、(+/−)-homo- nonactic acid (9/10)、2-methyl-2,5-bornandiol (11),其中化合物1为新化合物。 第三章主要进行研究云南美登木内生链霉菌Ly221的次生代谢成分,从中共分离得到5个化合物,分别是4,10-dihydroxy-10-methyl-dodec-2-en-1,4-olide (12)、两个非对映异构体4,11-dihydroxy-10-methyldodec-2-en-1,4-olides (13/14)、4-hydroxy-10-methyl-11-oxo-dodec-2-en-1,4-olide (15)和N-acetyltyramine(16)。 第四章主要研究Actinosynnema pretiosum ssp. aurantium ATCC 31565突变株HGF052的次生代谢成分,从中共分离得到7个化合物,分别是:N-demethyl- desepoxy-9-methoxy-maytansinol (17)、N-demethyl-desepoxy-maytansinol (18)、1-(furan-2-yl)ethane-1,2-diol (19)、两个混合物hexahydrofuro[3,2-b] furan-2,6-diol (20/21)。其中化合物17为新化合物,体外活性试验表明,该化合物对HL-60以及BEL-7402细胞株均有细胞毒活性,其IC50分别为0.12 μM和16.05 μM。 第五章综述了近十几年国内外关于植物内生放线菌的研究进展。
英文摘要: Endophytes are rich resources of novel natural products. In search for new bioactive secondary metabolites, we investigated chemical constituents of the fermentation extracts of three endosymbiotic Streptomyces strains and one mutant strain HGF052 derived from Actinosynnema pretiosum ssp. aurantium ATCC 31565. The thesis is composed of five chapters. Chapter 1 described the details on isolation, identification and bioassay of endophytes from Trewia nudiflora L. and Maytenus hookeri. In chapter 2, we investigated the secondary metabolites of Streptomyces sp. W5 and Streptomyces sp. M4, respectively. One new compound, cis-2-(1',8'-dihydroxy-8'- methylnonyl)-3-hydroxymethylbutanolide (1), together with nine known compounds, dimeric dinactin (3), (+/−)-nonactic acid (4/5), (R)-2-((2R,5S)-tetrahydro-5-(2- oxopropyl)furan-2-yl)propanoic acid (6), 2-(5-(2-acetoxypropyl)-tetrahydrofuran-2- yl)propanoic acid (7/8), (+/−)-homononactic acid (9/10), 2-methyl-2, 5-bornandiol (11) and an unidentified compound (2), were isolated. Their structures were elucidated on the basis of extensive spectroscopic data. In chapter 3, we reported the chemical constituents from the extract of fermentation medium of Streptomyces sp. Ly221. Five known compounds, 4,10-dihydroxy-10-methyl-dodec-2-en-1,4-olide (12), 4,11-dihydroxy-10-methyl- dodec-2-en-1,4-olides (13/14), 4-hydroxy-10-methyl-11-oxo-dodec-2-en-1,4-olide (15) and N-acetyltyramine (16), were isolated. In chapter 4, we studied on chemical constituents of a mutant strain HGF052 which derived from Actinosynnema pretiosum ssp. aurantium ATCC 31565. A new compound named N-demethyl-desepoxy-9-methoxy-maytansinol (17), and four known compounds, N-demethyl-desepoxy-maytansinol (18), 1-(furan-2-yl)ethane- 1,2-diol (19) and mixture of two compound hexahydro-furo[3,2-b]furan-2,6-diol (20/21), were isolated. Their structures were identified by means of spectroscopic analysis. In chapter 5, the author reviewed recent development in plant-originated endophytic actinomyces.
