题名: 川芎和当归抗ADP诱导血小板凝集活性成分研究

中文摘要: 本论文主要对川芎 (Ligusticum chuanxiong Hort.)和当归(Angelica sinensis (Oliv.) Diels)的化学成分和抗ADP诱导血小板凝集活性进行了研究。利用各种色谱分离技术(硅胶、硅胶H、Sephadex LH-20、Toyopearl HW-40、RP-8、RP-18),从上述两种植物中共分离得到了41个化合物,经波谱数据分析(UV, IR, MS, 1D, 2D-NMR)和X-单晶衍射分析,鉴定了5个新化合物和25个已知化合物:ligusticoside A (1),1, 2-二羟基-4-正戊基-3-环己烯 (4-pentylcyclohex-3-ene-1, 2- diol, 2),11R-myricanol (3),myricanone (4),12R- hydroxymricanone (5),洋川芎内酯 H (senkyunolide H, 6),洋川芎内酯 I (senkyunolide I, 7),洋川芎内酯 F (senkyunolide F, 8),4-羟基-3-丁基苯肽 (4-hydroxy-3-butylphthalide, 9),川芎三萜 (xiongterpene, 10),augustic acid (11),aromadendrane-4α,10β-diol (12), aromadendrane-4α,10α-diol (13),aromadendrane -4β,10β-diol (14),(4S)-p-menth-1- ene-4,7-diol (15),孕(甾)烯醇酮 (pregnenolone, 16),3 (R), 8 (S), 9 (Z)-falcarindiol (17),阿魏酸 (ferulic acid, 18),2-正丁基苯甲酸 (2-n-butylbenzoic acid, 19),4-羟基-3-甲氧基苯丙醇(4-hydroxy-3- methoxybenzenepropanol, 20),香草醛 (vanillin, 21),5-羟甲基糠醛 (5-hydroxymethyl-2-furancarboxaldehyde, 22),1, 3-二油酸-2-棕榈酸-甘油三酯 (1, 3-dioleate-2-palmitate -triglyceride, 23),4-O--D-呋喃阿洛酮糖--D-吡喃葡萄糖(4-O--D-furanoallulosyl--D-pyranoglucose, 24),β-谷甾醇 (β-sitosterol, 25),Z, Z'-(3.3', 8.8')-dibutylidenephthalide (26),Z, Z'-(3a.7a', 7a.3a' )-diligustilide (27),angelicolide A (28),正丁烯基苯酞 (Z-butylidenephthalide, 29),brefeldin A (30),其中化合物 1、2、26、27、28 为新化合物。通过活性筛选发现:在浓度为0.71 mg/ml时,化合物 2、4、5、6、7、9、11、17、19、22 共10个化合物具有一定抗ADP诱导血小板凝集活性。 论文最后综述了1979—2006年文献报道从川芎中分离得到的苯酞衍生物及其二聚体、酚类、有机酸类、生物碱类共计77个化合物,并对川芎主要的药理活性进行了概述。
英文摘要: This thesis focused on the chemical constituents and bioactivity on anti-platelet aggregation of Ligusticum chuanxiong and Angelica sinensis. 41 compounds were isolated from the two medicinal plants. Based on spectroscopic analyses (UV, IR, MS, 1D, 2D-NMR) and X-ray diffraction experiments, 30 compounds, including 5 new compounds, were elucidated as ligusticoside A (1), 4-pentylcyclohex-3-ene-1, 2-diol (2), 11R-myricanol (3), myricanone (4), 12R-hydroxymricanone (5), senkyunolide H (6), senkyunolide I (7), senkyunolide F (8), 4-hydroxy-3-butylphthalide (9), xiongterpene (10), augustic acid (11), aromadendrane-4α,10β-diol (12), aromadendrane-4α,10α-diol (13), aromadendrane -4β,10β-diol (14), (4S)-p-menth-1-ene-4,7-diol (15), pregnenolone (16), 3 (R), 8 (S), 9 (Z) - falcarindiol (17), ferulic acid (18), 2-n-butylbenzoic acid (19), 4-hydroxy-3-methoxybenzenepropanol (20), vanillin (21), 5-hydroxymethyl-2- furancarboxaldehyde (22), 1,3-dioleate-2-palmitate-triglyceride, (23), 4-O--D- furanoallulosyl--D-pyranoglucose (24), β-sitosterol (25), Z, Z'-(3.3', 8.8')- dibutylidenephthalide (26), Z, Z'-(3a.7a', 7a.3a' )-diligustilide (27), angelicolide A (28), Z-butylidenephthalide (29), brefeldin A (30). By anti-platelet aggregation assay, compounds 2, 4, 5, 6, 7, 9, 11, 17, 19, 22 presented potent anti-platelet aggregation activities at the concentration of 0.71 mg/ml. The last chapter summarized 77 chemical constitutes, such as phthalides, phthalide dimmers, organic acids, alkaloids, relevant to Ligusticum chuanxiong from 1979 to 2006, and summed up main physiological functions of Ligusticum chuanxiong.
