中文摘要: 本论文对四种药用及食用植物, 滇山茶 (Camellia reticulata Lindl.),白刺花(Sophora davidii (Fr.) Scheels),水茄(Solanum torvum Swartz.)和旋花茄(Solanum spirale Roxb.),进行了详细的化学成分研究,从中共分离鉴定了49个化合物,结构类型主要为多酚类和甾体配糖体,其中有12个新化合物。同时,对滇山茶花中的27个多酚类成分及7个白刺花进行了抗氧化活性评价,并综述了茄科植物的化学分类学研究和山茶属植物的化学成分及其生物活性。 第一章 滇山茶花的化学成分及其抗氧化活性研究 滇山茶(Camellia reticulata Lindl.),又名云南山茶,是山茶科山茶属著名的观赏花木,也是一种民间用药。本论文对滇山茶的鲜花进行了详细的化学成分研究,从中共分离鉴定了34个化合物,包括水解单宁5个,缩合单宁5个,黄酮类化合物16个,简单酚类成分8个。其中6个新化合物为:槲皮素3-O-[3-O--D-木糖吡喃糖基-6-O--L-鼠李吡喃糖基]--葡萄吡喃糖甙 (6),山萘酚 3-O-α-L-鼠李吡喃糖基-(1-3)-O-(2''', 4'''-O-二乙酰基)-α-L-鼠李吡喃糖基-(1-6)--D-葡萄吡喃糖甙 (22),山萘酚 3-O- (4''''-O-乙酰基)-α-L-鼠李吡喃糖基-(1-3)-O-(2''', 4'''-O-二乙酰基)-α-L-鼠李吡喃糖基-(1-6)--D-葡萄吡喃糖甙 (23),山萘酚 3-O-(3'''',4''''-O-二乙酰基)-α-L-鼠李吡喃糖基-(1-3)-O-(2''', 4'''- O-二乙酰基)-α-L-鼠李吡喃糖基-(1-6)--D-葡萄吡喃糖甙(24),山萘酚 3-O-(2'''',3'''',4''''-O-三乙酰基)-α-L-鼠李吡喃糖基-(1-3)-O- (4'''- O-乙酰基)-α-L-鼠李吡喃糖基-(1-6)--D-葡萄吡喃糖甙 (25),山奈酚3-O-(2'''',3'''',4''''- O-三乙酰基)-α-L-鼠李吡喃糖基(1-3)- (3'''-4'''- O-二乙酰基)-α-L-鼠李吡喃糖基(1-6)- -D-葡萄吡喃糖甙 (26)。抗氧化活性研究结果表明,大多数多酚类化合物都表现出较强的消除自由基能力。 第二章 白刺花根的化学成分及其抗氧化活性研究 白刺花(Sophora davidii (Fr.) Scheels)为豆科槐属植物,其根、茎、叶和花均为民间药物,用于治疗咽喉肿痛、肺热咳嗽、肝炎、痢疾、淋症、水肿、衄血、便血、血尿等病症。 本论文对采自云南省文山州的白刺花根进行了化学成分研究,从中分离到了8个化合物,包括3个异黄酮类和4个二苯乙烯类化合物。已经鉴定了7个化合物的结构,分别为芒柄花黄素(1),Davidiol B (2),Davidiol E (3),三叶豆紫檀甙 (4),高丽槐素 3-O-α-L-木吡喃糖基-(1-6)--D-葡萄吡喃糖甙 (5),白藜芦醇 (6),Davidiol F (7)。其中化合物3,5和7为新化合物。对已经鉴定结构的7个化合物进行了抗氧化活性评价,只有化合物 6表现出较好的的抗氧化活性。 第三章 两种茄属食用植物的主要化学成分研究 水茄(Solanum torvum Swartz.)的果实是当地常见的野生蔬菜。民间还用其根治疗跌打瘀痛、腰肌劳损、胃痛、牙痛、闭经、久咳等。本论文对西双版纳产水茄的新鲜果实主要化学成分进行了研究,从中分离得到4个甾体配糖体,26-O--D-葡萄吡喃糖基-(25S)-5α-呋甾-22ξ-甲氧基-6α,26-二醇-3-酮-6-O-α-L-鼠李吡喃糖基(1→3)--D-奎纳吡喃糖甙 (22-methoxyl torvoside H) (1),26-O--D-葡萄吡喃糖基-(25S)-5α-呋甾-22-甲氧基-3,6α,26-三醇-6-O-α-L-鼠李吡喃糖基(1→3)--D-奎纳吡喃糖甙 (22-methoxyl torvoside A) (2),26-O--D-葡萄吡喃糖基-(25S)-5α-呋甾-22-甲氧基-6α, 26-二醇-3-酮-6-O--D-奎纳吡喃糖甙 (torvoside E) (3),(22R,23S,25R)-螺甾-5α-3,6α,23-三醇-6-O-α-L-鼠李吡喃糖基(1→3)--D-奎纳吡喃糖甙(torvoside K) (4),其中化合物1和2为新化合物。 旋花茄(Solanum spirale Roxb.)的嫩叶和果实为我国西双版纳傣族等少数民族的常用蔬菜,还是民间常用药物,印度用根作为麻醉剂和利尿剂,越南用叶治疗脚气和胃肿胀。本论文从旋花茄的新鲜果实的甲醇提取物中分离鉴定了3个甾体皂甙,26-O-β-D-葡萄吡喃糖基-呋甾-(25R)-22ξ-甲氧基-3β,26-二醇-5-烯-3-O-α- L-鼠李吡喃糖基(1-2)-β-D-葡萄吡喃糖甙 (proto-type of prosapogenin A of dioscin) (5),26-O-β-D-葡萄吡喃糖基-呋甾-(25R)-3β,22ξ,26-三醇-5-烯3-O-α-L-鼠李吡喃糖基(1-2)-[α-L-鼠李吡喃糖基(1-4)]-β-D-葡萄吡喃糖甙 (protodioscin) (6),26-O-β-D-葡萄吡喃糖基-呋甾-(25R)-3,22ξ,26-三醇-5-烯-3′′-乙酰基-3-O-α-L-鼠李吡喃糖基(1-2)[α-L-鼠李吡喃糖基(1-4)]-β-D-葡萄吡喃糖甙(7),其中化合物7是新化合物。 第四章: 本文主要对茄科植物的化学分类学研究进行了综述。 第五章: 本文对80年代以来,国内外发表的山茶属植物的化学成分及药理作用研究进行了综述。
英文摘要: The phytochemical investigations on four medicinal and edible plants in Yunnan province, including Camellia reticulata Lindl., Sophora davidii (Fr.) Scheels, Solanum torvum Swartz., and S. spirale Roxb., led to the isolation of forty-nine polyphenolics and steroidal glycosides. Twelve of them are new compounds. Most of the compounds isolated from C. reticulata and S. davidii were tested for their aintioxidant activities. In addition, the chemtaxonomy of Solanaceae and the advances including chemistry and biological activities of genus of Camellia after 1980’s were also reviewed. This thesis consists of the following parts: Chapter 1. Studies on the chemical constituents from the fresh flowers of Camellia reticulata and their antioxidant activities Camellia reticulata, also called “Yunnan Shancha”, is a famous Camelliaceous ornamental plant. It is also used as a folk medicine for the treatment of haematemesis, diarrhea, and scald,. Thirty-four phenolic constituents, including 5 hydrolysable tannins, 5 condensed tannins, 16 flavonoids and 8 simple phenolics were isolated from the fresh flowers of this plant. Of these, six compounds were new flavonoids: quercetin 3-O-[-D-xylopyranosyl(1-3)]--L-rhamnopyranosyl(1-6)--glucopyrano- side (6), kaempferol 3-O-α-L-rhamnopyranosyl-(1-3)-(2''',4'''-O-diacetyl)-α-L- rhamnopyranosyl-(1-6)--D-glucopyranoside] (22), kaempferol 3-O-(4''''-O-acetyl)- α-L-rhamnopyranosyl-(1-3)-(2''',4'''-O-acetyl)-α-L-rhamnopyrnsyl-(1-6)--D-gluco pyranoside] (23), kaempferol 3-O-(3'''',4''''-O- diacetyl)-α-L-rhamnopyranosyl-(1-3) - (2''',4'''-O-diacetyl)-α-L-rhamnopyranosyl (1-6) --D-glucopyranoside] (24), kaemp- ferol 3-O-(2'''',3'''',4''''-O-triacetyl)-α-L-rhamno pyranosyl-(1-3)-4'''-O-acetyl)-α- L- rhamnopyranosyl-(1-6)--D-glucopyranoside] (25), kaempferol 3-O- (2'''',3'''',4''''- O-triacetyl)-α-L-rhamnopyranosyl-(1-3)-(2''',4'''-O- diacetyl)-α-L-rhamnopyranosyl- (1-6)--D-glucopyranoside] (26). Antioxidant testing showed that most of polyphenolics displayed significant DPPH radical scavenging activities. Chapter 2. Studies on the chemical constituents from the roots of Sophora davidii and their antioxidant activities. Sophora davidii, a Leguminous folk medicine, has been used to treat hepatitis, diarrhea, edema. Eight compounds have been isolated from the roots collected from Wenshan county of Yunnan province. Seven of them were determined to be formonoetin (1), Davidiol B (2), Davidiol E (3), trifolorhizin (4), Maackiain 3-O-b-D- xylopyranosyl-(1-6)-b-D-glucopyranoside (5), resveratrol (6), and Davidiol F (7) . Compounds 3, 5 and 7 are new constituents. Antioxidant testing showed that only compound 6 exhibited remarkable antioxidant activity, while others presented moderate activities. Chapter 3 Studies on the chemical constituents of two edible Solanum plants Fruits and leaves of Solanum torvum and S. spirale are used as vegetable in Xishuangbanna Yunnan province. A decoction of Solanum torvum fruits is considered useful in cases of liver and spleen enlargement. Our chemical study on its fresh fruits resulted in the isolation of four major constituents, which were assigned as steroidal saponins, 22-methoxyl torvoside H (1), 22-methoxyl torvoside A (2), torvoside E (3), and torvoside K (4). Among them, compounds 1 and 2 are new steroidal glycosides. S. spirale, as a folk medicine, can cure beriberi, swollen stomach, narcotic and diuretic diseases. Three furostanol saponins, proto-type of prosapogenin A of dioscin (5), protodioscin (6), and 3′′-acetyl-protodioscin (7), were isolated from the fresh fruits of this plant. Of them, saponin 7 having an acetyl group attched at the sugar moiety is a new steroidal glycosides. Chapter 4 The chemtaxonomy of Solanaceae have been reviewed. Chapter 5 The advances including chemistry and biological activities of genus of Camellia after 1980’s were reviewed.
