瘤枝五味子的化学成分及其生物活性研究

摘要瘤枝五味子(Schisandra bicolor var. tuberculata)为木质藤本植物,隶属于五味子科五味子属,是二色五味子(S. bicolor)的变种,主要分布于贵州、湖南、江西等地区。其根和茎有理气活络、健脾之效,可缓解过度劳累、四肢酸麻、食欲不佳、胸闷等症。前人对产自湖南新宁的瘤枝五味子,以及产自浙江天目山和江西上饶的二色五味子进行过化学成分及其生物活性的研究,结果表明,二色五味子以及变种瘤枝五味子含有木脂素、三萜、倍半萜等化学成分,部分化合物呈现良好的生物活性。鉴于前人对瘤枝五味子的研究较少,为了加深对该种植物化学成分的认识、丰富本研究组的化合物库及探索不同产地瘤枝五味子次生代谢产物的差异,本研究对产自贵州铜仁的瘤枝五味子藤茎部位进行了系统的化学成分研究。 本论文通过各种色谱分离技术、现代波谱学方法、单晶X射线衍射以及量子化学计算等方法,从上述植物中分离鉴定了53个化合物,其中新化合物16个。所得化合物中,包括木脂素41个,(新化合物14个);三萜4个;杜松烷型倍半萜4个(新化合物2个);以及其他类型化合物4个。本论文成功实现了5对对映异构体的手性拆分;通过单晶X射线衍射结合量子化学计算ECD确定了化合(?)-15的绝对构型;通过量子化学计算NMR参数确定了化合物(?)-22、38、46和47的相对构型。对分离得到的部分化合物进行了神经保护活性的筛选,其中倍半萜类化合物48和49对皮质酮诱导的PC12神经细胞损伤具有一定的保护作用。通过进一步分析本研究内容与前人对瘤枝五味子的发现表明:该种植物的主要化学成分为木脂素类化合物,且产地差异对植物中所含有的主要木脂素结构类型产生较大影响。; Schisandra bicolor var. tuberculata (Y. W. Law) Y. W. Law is a climbing woody liana belonging to the genus Schisandra (Schisandraceae), and it is mainly distributed in Guizhou, Hunan, Jiangxi and other regions. Its roots and stems can be used to alleviate overstrain, numbness of limbs, poor appetite, chest tightness, etc. Previous studies on chemical constituents and biological activities of S. bicolor var. tuberculata from Xinning, Hunan Province, and S. bicolor from Tianmu Mountain, Zhejiang Province and Shangrao, Jiangxi Province have demonstrated that S. bicolor and S. bicolor var. tuberculata contained various compounds, such as lignans, triterpenoids and sesquiterpenoids, and some of them displayed good biological activities. Considering that S. bicolor var. tuberculata has rarely been phytochemically investigated, in the present study, the stems of S. bicolor var. tuberculata from Tongren, Guizhou were subjected to phytochemical research, with the purpose of mining more intriguing chemical constituents from this plant, enriching the compound library in our laboratory, and exploring the differences of chemical compositions among S. bicolor var. tuberculata from different habitats. By means of various chromatographic and spectroscopic techniques, quantum chemical calculations, as well as single-crystal X-ray diffraction analysis, 53 compounds, including 16 new ones, were isolated and identified from S. bicolor var. tuberculata. These compounds consisted of 41 lignans (14 new ones), 4 triterpenoids, 4 cadinane-type sesquiterpenoids (2 new ones), and 4 other miscellaneous compounds. In this thesis, chiral resolution of 5 pair of lignan enantiomers was completed. The absolute configurations of (?)-15 were determined by single-crystal X-ray diffraction analysis and quantum chemical calculation of electronic circular dichroism (ECD) spectrum. The relative configurations of compounds (?)-22, 38, 46 and 47 were determined by quantum chemical calculation of nuclear magnetic resonance (NMR) parameters. A few compounds were evaluated for their neuroprotective activity, and two sesquiterpenoids, 48 and 49 exhibited moderate protective activities against corticosterone-induced apoptosis in PC12 cells. In combination with previous studies on S. bicolor var. tuberculata, it can be found that the main chemical constituents of this plant are lignans, and the difference in growing environments have a great influence on the structural types of lignans produced by the plant.
