松叶蕨和翠云草化学成分研究

摘要本论文分为两章,分别以松叶蕨(Psilotum nudum (L.) Beauv)和翠云草 (Selaginella uncinata (Desv.) Spring)两个蕨类植物为研究对象,论述了其化学 成分及细胞毒和抗炎活性研究。通过系统研究,从两种蕨类植物中共分离鉴定 40 个化合物,6 个新化合物,并对分离得到的新化合物和双黄酮类化合物进行 了 5 株肿瘤细胞的体外细胞毒活性和 NO生成抑制活性筛选。 第一章论述了松叶蕨的化学成分及生物活性研究。松叶蕨为松叶蕨科 (Psilotaceae)松叶蕨属(Psilotum)植物,是古代孑遗种,也是蕨类植物中的珍稀濒 危植物。松叶蕨可全草入药,作为民族药治疗风湿关节痛,女性经闭,吐血及 外伤性损伤。由于其资源稀缺,相关研究鲜有报道。本研究从松叶蕨的 300 g 干燥全草中共分离鉴定 24 个化合物,结构类型包括双黄酮类,酚类和长链脂肪 酸类化合物,其中 6 个新化合物。并对分离得到的新化合物和双黄酮类化合物 进行了五株肿瘤细胞的体外细胞毒活性和 NO 生成抑制活性筛选 ,根据筛选结 果可知,化合物 1 对于 HL-60、SMMC-7721 和 A549 细胞有一定的抗增殖作 用。IC 50 值分别为 13.28 ± 1.73,17.82 ± 0.53 和 32.90 ± 0.84 μM。此外 5 个化合 物具有不同程度的抗炎活性,化合物 6 活性最显著,IC 50 值为 7.22 ± 0.23 μM。 第二章叙述了翠云草的化学成分及生物活性研究。翠云草为卷柏科 (Selaginellaceae)卷柏属(Selaginella)植物,为中国特有。其外形美观易栽培,具 有园林观赏价值,同时也作为民族药而被广泛使用,具有清热利湿,止血,治 疗关节疼痛的功效。本研究综合利用各种色谱技术,波谱分析等手段,从翠云 草的干燥全草中共分离鉴定了 16 个单体化合物,结构类型包括双黄酮类,酚 类,萜类,生物碱类,和长链脂肪酸类。同时对 4 个双黄酮类化合物进行了细 胞毒和抗炎活性筛选,结果均未显示有明显活性。; The dissertation consists of two chapters , which reported the chemical constituents and their cytotoxic and anti-inflammatory of Psilotum nudum (L.) Beauv and Selaginella uncinata (Desv.) Spring, respectively. Through various chromatographic methods and spectral analysis technology, 40 compounds were isolated from the herbs of two pteridophytes, including six new compounds. In addition, all the new compounds and biflavonoids were screened for their cytotoxic and NO production inhibition activity in vitro. The first chapter described the study of chemical constituents and their bioactivity of P. nudum (L.) Beauv. It is a plant of the genus Psilotum in the family Psilotaceae, and is an ancient relic species and a rare endangered plant among ferns. It also can be used as medicine for rheumatism arthralgia, amenorrhea, hematemesis and traumatic injury. Due to scarce resources, there are few reports about the chemical components of P. nudum (L.) Beauv. In this study, a total of 24 compounds were isolated from the dried whole grass of P. nudum (L.) Beauv, six of which were new compounds. The structural types of other known compounds include biflavonoids, phenols and fatty acids. Some of the isolated compounds were tested for their cytotoxic and NO production inhibition activity in vitro. As a result, compound 1 could inhibit the proliferation of cell HL-60, SMMC-7721and A549, the IC 50 values were 13.28 ± 1.73, 17.82 ± 0.53 and 32.90 ± 0.84 μM, respectively. In addition, five isolated compounds showed certain inhibitory effects on NO production, and compound 6 showed significant anti-inflammatory activity with IC 50 value of 7.22 ±0.23 μM. The second chapter focused on research of the chemical constituents from S. uncinata (Desv.) Spring. and their cytotoxic and NO production inhibition activity. Selaginaceae is widely distributed, while S. uncinata (Desv.) Spring is native to China. It is so beautiful in appearance and easy to cultivate that it has ornamental value in gardens. S. uncinata (Desv.) Spring is also used as a national medicine to clear heat and promote diuresis, stanch bleeding and treat arthralgia of muscle and bone. In this study, On the basis of a wide range of isolation and elucidation techniques, 16 known compounds were isolated and identified from S. uncinata (Desv.) Spring. The classes of these compounds include biflavone, phenols, sesquiterpenoids, alkaloids and long-chain fatty acids. In addition, the screening results of cytotoxic and anti-inflammatory activities of the four isolated biflavonoids showed no activity.
