摘要 | Natural products are the source of drug Discovery, and the direct source of therapeutic leads. Yunnan is one of the regions with the richest biodiversity in the world, with many relict species, endemic species and rare species, 125 of which are endemic to China, accounting for 52.3% of endemic genera in China. Endemic species bred characteristic secondary metabolites, which are valuable resources for research on undescribed and active ingredients This thesis mainly introduces the chemical constituents and biological activities of two Yunnan peculiar plants: Stemona mairei Krause and Bousigonia angustifolia Pierre. A total of 209 compounds, including 74 new ones, were isolated from the above two plants based on the many separation technics and spectrometric analysis, single crystal X-ray diffraction and quantum chemical calculation of NMR paprameters. Plant of S. mairei in the Stemonaceae family are rich in alkaloids and stilbene compounds. 82 compounds, including 37 new ones were isolated and elucidated from the roots of S. mairei. The first dimer of Stemona alkaloids was discovered, the molecule is connected by a pyridine ring in a symmetrical way. It may be formed by oxidation, degradation, and rearrangement of the Protostemonine monomer. This discovery have changed the "common sense" of the simplex structure of Stemona alkaloids and enriched the skeleton types of Stemona alkaloids. The results of activity studies show that Stemona alkaloids not only have traditional insecticidal activity, but also have hypoglycemic activity for the first time. Plant of B. angustifolia in the apocynaceae family are rich in monoterpenoid indole alkaloids. In this study, 127 compounds, including 37 new ones were isolated and elucidated from its stems and leaves of B. angustifolia. Compounds 1b, 2b and 3b are all novel skeletons of polymeric monoterpene indole alkaloids, of which 1b is the first symmetrical monoterpene indole Alkaloid trimer. At the same time, the biogenic precursors of 1b, 2b and 3b were also discovered, which supports our speculation on the biogenic pathways of the three compounds. The tumor cytotoxicity of these alkaloids was evaluated which indicated that compound 39b has a significant inhibitory effect on SMMC-7721, Hela and HepG2, with IC50 values of 0.520, 0.600 and 0.758μM, respectively. |
