药用植物资源组分库的构建及其抗菌活性评价; Construction and antimicrobe screening of library of extract fractions from medicine plants

摘要Natural products are one of the important sources for the innovation of new drugs, whereas the activity evaluation for the crude extracts and the confirmation of bioactive fractions of medicinal plants are extremely crucial for the further discovery of the lead compounds. Construction of a fraction library for medicinal plant resources could be suitable for the further researches of medicinal plant resources with great variety and quantity, which could greatly promote their basic research process and it possesses great significance to improve the development and utilization for medicinal plant resources. The establishment of a fraction entity library can not only enable it become an important reserve resource, but also provide a variety of fraction research data, which provide indispensable theoretical support for their subsequent development and utilization. The dissertation aims to quickly and systematically construct the fractions library of medicine plant resources by means of creating standard procedure. This was achieved by standard extraction and prefractionation to obtain eight hundred subfractions of one hundred medicinal species. In addition, the components of these subfractions were preliminarily characterized by TLC, and the results of TLC and the quality data of all the fractions were detailed obtained, and summarized the research summary of the one hundred medicinal plants, including the main chemical constituents and their pharmacological effects, can serve as a theoretical basis for the further research and development of medicinal plant resources. In addition, the antibacterial activity evaluation of the medicinal plant resource fraction library has been carried out. Explained how to obtain active fractions by a combined approach of the fraction library of medicinal plant resources and a high-efficiency screening method. The MTT colorimetric method was used in this study to determine the antibacterial of aforementioned eight hundred subfractions. The results revealed that fourteen fractions showed antimicrobial effect (MICs ≤ 50.0 μg/mL), which were superior to those in current reported literature. Among them, two fractions exhibited bactericidal activity (MBCs = 25.0 μg/mL) against Staphylococcus aureus, and their MIC and MBC values are lower than the positive control ampicillin (MICs = 50.0 μg/mL , MBCs > 50.0 μg/mL). There fractions also displayed a certain antibacterial effect on the five tested bacteria. The above results laid a solid foundation for the research of effective antibacterial components in the future. In addition, thirty fractions were disclosed to showed antimicrobial activities against three plant pathogenic fungi (MICs ≤ 50.0 μg/mL). Four of them displayed remarkable antifungal activities against Botrytis cinerea at a concentration of 12.5 μg/mL, and their anti-fungal effects were stronger than those of positive control drugs carbendazim and chlorothalonil (MFCs > 50.0 μg/mL).
