小白及菲类成分及其应用探究; Phenanthrene components of Bletilla formosana and its applied exploration

摘要Bletilla genus, a kind of Orchidaceae plants, can be frequently seen in sub-tropical and tropical area, and the tubers of this genus has been used as an astringent hemostatic, detumescence and promoting granulation. In recent years, hemostasis and anti-inflammatory activity of Bletilla have witnessed extensive applications in the field of drug development. This dissertation consists of two chapters. Based on the literature research on the chemical constituents of Bletilla genus, the extraction and separation work of B. formosana was carried out with the aim of exploring the differences in chemical constituents between diverse species of Bletilla and more importantly, discover new compounds. the application of the isolated monomer compounds was explored, which provided theoretical basis for the drug development of B. formosana. The first chapter summarizes the progress in the chemical constituents and pharmacological activities of Bletilla genus, which provided a reference for the extraction and isolation of B. formosana and the preliminary study of its biological activity. The second chapter studies preliminarily the phenanthrene components of the secondary metabolites of Bletilla formosana (Hayata) Schltr., which is one of the species of Bletilla genus. A variety of chromatographic separation techniques such as silica gel column chromatography, Sephadex LH-20 and semipreparative HPLC were used to separate and purify compounds, and their structures were identified on the basis of ultraviolet, infrared, mass spectrometry and extensive spectroscopic methods. 39 compounds were isolated and identified from B. formosana, including 2 new compounds, 1,5-bis(p-hydroxybenzyl)-4,7-dimethoxy-9,10-dihydrophenanthrene-2,8-diol (1*) and 4-(p-hydroxybenzyl)-2,5-dimethoxy-9,10-dihygrophenanthrene-1,7-diol (2*). Among them, compounds 3-22, 25-38 were isolated and purified from B. formosana for the first time. Phenanthrene was the main structural type of the compounds, and the others were phenolics, acetaldehyde, glucosyloxybenzyl 2-isobutylmalate and steroids. Traditional applications based on Bletilla, Phenanthrene compounds isolated from B. formosana were screened for nitric oxide activity, anti-inflammatory activity and antioxidant activity. The result reveals, sixteen compounds showed significant DPPH radicals scavenging activity; Eleven compounds showed inhibition effects on the NO production in LPS-stimulated RAW 264.7 cell, among which compound 1, 3, 4, 11, 13 showed significant inhibition effects; Fifteen compounds had some antitumor cytotoxic activity, among which compound 3 was the best (IC50 4.59~13.98 μM). This study provides scientific basis for further development and utilization of B. formosana resources, which has certain theoretical significance and potential application value.
