摘要 | The Dendrobium species are one of genus in the Orchidaceae family with both ornamental value and medicinal value. This genus has the effects of nourishing body, protecting the stomach and promoting the production of body liquid. The ancient prescription also records that Dendrobium species can be used for the treatment of "Xiaoke" disease. As the main component in Dendrobium species, polysaccharides are considered to be the key reason for its “Xiaoke” effects, so it has attracted great attention from scholars. This dissertation is composed of four chapters. The first chapter is a review of the progress of the structure and activity of polysaccharides from Dendrobium species, the progress of technology in extraction, separation and identification of polysaccharides, and the progress of diabetes drugs and hypoglycemic substances based on the GLP-1 secretion. The second chapter describes the structural characterization of polysaccharides from four Dendrobium species and their GLP-1 secretion-promoting activity. The third chapter describes the structural characterization of polysaccharides from D. officinale and their hypoglycemic activity and mechanism based on GLP-1 pathway. The fourth chapter is the structural characterization of polysaccharides from D. devonianum and their GLP-1 secretion-promoting activity and mechanism. Chapter 1 Summary Firstly, this chapter reviews the structure and pharmacological activities of polysaccharides reported in Dendrobium species, and provides a basis for the discovery of the structure-activity relationship of polysaccharides from Dendrobium species. Then, the progress of technology in extraction, separation and identification of polysaccharide is summarized, which is benefit to solve various problems in preparation of polysaccharides. Finally, a report of diabetes drugs and hypoglycemic substances that can stimulate GLP-1 secretion has been given. This attempt is to find a leading drug for promoting GLP-1 secretion Chapter 2 The structural characterization of polysaccharides from four Dendrobium species and their GLP-1 secretion-promoting activity This chapter describes the structural characterization and GLP-1 secretion-promoting activity of polysaccharides from D. officinale, D. devonianum, D. nobile and D. aphyllum. It is found that the four kinds of polysaccharides differ greatly in their main molecular weight, sugar content, monosaccharide composition and ratio. The ability of promoting GLP-1 secretion is also different, which may be caused by the difference in their structure. Therefore, in order to find out the relationship of structure-activity, it is important to get more precise information of structural characterization of each polysaccharide.
