摘要 | Abstract Salvia, one of the largest genera (about 1000 species) of the widespread family Lamiaceae, is distributed worldwide in the temperate and warm zones. Many species of this genus are used as folk medicine to treat a wide variety of ailments throughout the world. The genus Salvia is a rich source of diterpenoids with structural diversity. Many diterpenoids with attractive bioactivities, such as tanshinone IIA, salvicine, neotanshinlactone, and salvinorin A, have been reported from this genus. As the polyphenolic acid B and tanshinone IIA (sodium sulfonate) are clinically used for the treatment of coronary heart disease, angina pectoris, and myocardial infarction. Diterpenoids with abietane skeletons is considered as the main bioactive and characteristic chemical constituents of this genus. Abietane diterpenoids usually have many conjugated chromophores. Expressly, different kinds of abietane have typical UV absorptions. In addition, a little change of architecture will lead the bathochroic or hypsochromic shift of UV. As a part of our ongoing research about the plants of Salvia species, an oriented isolation and purification method of nature abietane diterpenoid have established by the characteristic UV absorptions in HPLC. Our group has been committed into the phytochemical studies of genus Salvia more than 10 years, and has discovered a series of novel terpenoids with diverse structures and bioactivities. The phytochemical study on Salvia digitaloides led the isolations of 55 diterpenoids (nine of which are new compounds, including one featuring novel carbon skeleton with 6/7/6 ring system and unique C10-methylation) with varieties of abietane, icetexane, labane and pimarane. Notably, three (including two new ones) rare sempervirane-type diterpenoids were also obtained. Unexpectedly, three new isoprenylated flavonoids with various flavanone, pterocarpan, and isoflavanone architectures were isolated from S. digitaloides. It is first time that isoprenylated flavonoids were reported from Salvia. In this study, the antiplatelet activity of the obtained abietane diterpenes was screened, which showed 24 could significantly reverse platelet aggregation. Interestingly, 7 and 21 could induce platelet aggregation. In addition, all the isoprenylated flavonoids were evaluated for their activities on Cav3.1 low voltage-gated Ca2+ channel (LVGCC), of which 57 strongly and dose-dependently inhibited Cav3.1 peak current. In summary, these findings could enrich diversities of secondary metabolites of the genus Salvia, increase universality of biological activities and properties, and expand the research contents of chemical constituents and biological activities of Salvia. |
