飞龙掌血化学成分及细胞毒活性研究; Studies on Chemical Constituents and Cytotoxic Activity of Toddalia asiatica

摘要In this thesis, chemical investigation was carried out with the dried roots of medicinal plant Toddalia asiatica and 30 compounds were obtained and structurally assigned as 14 alkaloids (1–14), 13 coumarins (15–27), one polyketide (28) and two phenolic acids (29–30). Their structures were identified as desformyl-isoarnottianamide (1*), dihydrochelerythrine (2), 8-acetonyldihydrochelerythrine (3), decarine (4), zanthoxyline dimethoxy derivative (5), noravicine (6), N-nornitidine (7), oxyavicine (8), oxynitidine (9), skimmianine (10), haplopine (11), γ-fagarine (12), dictamnine (13), N-methylflindersine (14), toddalenone (15), toddalolactone (16), limettin (17), coumurrayin (18), 5-methoxyseselin (19), braylin (20), toddanone (21), omphamurin (22), toddaculin (23), O-methylcedrelopsin (24), 7-geranyloxy-6-hydroxycoumarin (25), 7-geranyloxy-6-methoxycoumarin (26), 8-geranyloxy-5,7-dimethoxycoumarin (27), penialidin C (28), vanillin (29), and 4-hydroxybenzaldehyde (30). Among them, 1* was identified as a new compound, and 25 as a new natural product. Compound 28 was firstly isolated from plants, and 26 from the family Rutaceae. Compounds 5–7 were obtained from the plants of genus Toddalia for the first time. The results of cytotoxic assay revealed that compounds 2, 4, and 28 showed significant antiproliferative effects on human lung carcinoma cell line H1299-EGFP with IC50 values ranging from 4.2 to 5.3 μg/mL (Positive control CPT-11, IC50 = 1.1 μg/mL), whereas 2, 3, 4, 27 and 28 exhibited remarkable antiproliferative activities against human hepatic carcinoma cell line MHCC97H-EGFP with IC50 values ranging from 1.0 to 7.3 μg/mL, equivalent to that of CPT-11 (IC50 = 5.5 μg/mL). The specific mechanism of action for the abovementioned active ingredients remains to be further studied. Additionally, the distribution of the obtained alkaloids from T. asiatica was analyzed via network analysis technology, and the results suggested a close relationship between Toddalia and Zanthoxylum, as well as relations between Toddalia and other various genera of different families. These findings supplied lots of underlying and helpful information for taxonomic researches from a phytochemical perspective, and also provided guidance for phytochemists to hunt more alkaloids with similar structures from the related plants.
