滇重楼中萜类生源天然产物的生物合成研究; Biosynthesis of natural terpenoids from Paris polyphylla var. yunnanensis

摘要Terpenoids, also known as isoprenes, are the largest class of natural products with diverse chemical structures and biological activities. Paris polyphylla var. yunnanensis is a famous Chinese herbal medicine, which is the important raw material of "Yunnan Baiyao", "Gongxuening Capsule" and other Chinese patent medicines. The main active ingredients of P. polyphylla var. yunnanensis are steroidal saponins, which originate from the isoprene unit and also belong to broad definition of terpenoids. The key enzymes involved in the upstream biosynthetic pathway of terpenoids and downstream biosynthetic pathway of steroidal saponins in P. polyphylla var. yunnanensis were studied, including isoprenyl diphosphate synthases (prenyltransferases), terpenoid synthases and cytochrome P450 monooxygenases (CYP450s). Two candidate prenyltransferase genes, three candidate terpenoid synthase genes and 62 candidate CYP450 genes were cloned from P. polyphylla var. yunnanensis. The functions of one squalene synthase and one GGDP synthase were identified by in vivo and in vitro enzymatic activity assay. The expression pattern of the squalene synthase gene was found to be related to the accumulation profiles of Paris saponins. In addition, engineered Saccharomyces cerevisiae producing cholesterol and diosgenin were constructed, which laid a foundation for further function characterization of candidate CYP450 genes involved in biosynthesis of Paris saponins.
