摘要Medicinal plant Paris polyphylla Smith var. yunnanensis (Franch.) Hand. -Mazz. belongs to the Melanthiaceae family. Its rhizome can be used as medicine, and it is widely used as traditional Chinese medicine. Polyphyllins, the active ingredient of P. polyphylla var. yunnanensis, have the effective analgesic, anti-inflammatory, hemostasis, inhibition of cancer cell and proliferation actions. Polyphylla var. yunnanensis is a perennial geophyte plant, the rhizome of which is a underground storage organ and the aboveground part is annual. The growth of P. polyphylla var. yunnanensis is coincident with the wet and dry seson in Yunnan Province. As an important raw material for pharmaceutical industry and clinical medicine, the P. polyphylla var. yunnanensis rhizome has important biological significance and socio-economic value. But it is still in the transition stage from wild to domestic species. P. polyphylla var. yunnanensis has a large and complex genome and a long development cycle. And the variation brought by the interspecific hybridization has made it technically difficult to study the physiological mechanism. The relevant knowledge is almost blank. In this paper, we first observed the morphological and structural changes in different stages of the development of P. polyphylla var. yunnanensis buds. Then, the effects of hormones and environmental temperature cycles on bud morphological development, were explored. At the last, the regulation mechanism of hormones on dormancy, and the bud differentiation and dormancy regulation from morphological anatomy, hormone regulation and environmental response were analyzed. The main findings are as follows: We use the in vitro system of somatic embryo regeneration plants as the research system. Through the observation and dissection buds in vitro, we found that P. polyphylla var. yunnanensis development process can be divided into five stages, namely the seedling rhizome formation apical bud differentiation dormancy and growth. The rhizome was formed and the buds and only experienced differentiated once during first three stages, while it will experience dormancy and growth repeatedly afterwards. In the rhizome of P. polyphylla var. yunnanensis, the apical buds will first developed and many axillary buds grow near the apical buds in the same year. When the apical buds begin to live on the ground in the first year, the axillary buds live underground and gradually complete the differentiation of the stems and leaves, which is ready to become a new apical buds in the next year. Gibberellin induces polyaerial shoot formation and increases the propagation rate in P. polyphylla var. yunnanensis. We found that 20 mg/L GA3 could break dormancy and induced polyapical shoots. The apical shoot number per GA3-treated rhizome reached 7.2 per stem. The GA3-induced occurrence of polyapical shoots derived from the repetitive de novo formation of apical buds.
