滇西北三种垫状植物土壤微生物空间异质性分布的研究; Study on the spatial heterogeneity distribution of microbial community of three cushion species in NW Yunnan, SW China

摘要Alpine cushion plant is commonly referred to plant species sharing hemispheric aboveground structure occurred at high elevations. As one of the most pronounced example of the highly specialized morphology occurred among systematically distant plant species at alpine areas, cushion species is well documented for their outstanding capacity on modifications of environmental conditions, in particular soil water and nutrients. Recently, following the progresses on sequencing technology in molecular biology study, the impact of the modifications of soil environments by cushion plant on microbial community diversity, composition and structure has been gradually unveiled. Yet, the spatial distributions of microbial community and its functional groups among different plant species sharing similar cushion morphology and their correlations with above- and belowground plant traits and soil chemical characteristics are largely remained unknown. In this study, we selected three typical cushion plant species (Androsace delavayi, Arenaria polytrichoides, Rhodiola coccinea var. scabrida) inhabit at alpine screes of Baima Snow Mountain, Deqen County, Northwest Yunnan Province and examined the microbial community of soil samples collected at four different spatial locations therein: inside and at the edge of aboveground cushion structure, rhizosphere and surrounding bare ground uncovered by any cushion species using high-throughput sequencing technology. The main findings are as follows: (1) There were significant differences in microbial community richness (observed species) and diversity (Shannon index) observed at the same spatial location among different cushion species, while no significant spatial difference was found for each of them. Of the three cushion species studied, R. coccinea var. scabrida exhibited the greatest microbial community richness at all four spatial locations, while the microbial community richness and diversity at rhizosphere of A. delavayi were the lowest. (2) With the increases of soil pH value and root length, the richness and diversity of soil microbial community increased. (3) The microbial community exhibited clear species-specific and spatial difference in their compositions, dominant taxon and functional groups, with a great proportion of some nitrogen cycling related groups (e.g. nitrification, denitrification and nitrate reduction bacteria). This suggested a potential facilitation from microbes on nutrient absorption for cushion plant growing on alpine screes, which is a typical poor nutrient habitat. By evidencing a pronounced species-specific and spatial difference in microbial community among different cushion species and highlighting the importance of abiotic (soil pH) and biotic (root length and/or cushion structure) factors in determination, our study provides some new information that improves the understanding of plant-microbe interactions at high elevations and their ecology significance on the adaptation of cushion
