摘要 | Obesity is one of the most prevalent public health concerns worldwide, and plenty of evidence has proved that obesity serves as an important trigger for a variety of diseases such as insulin resistance (IR), type 2 diabetes (T2D), hyperlipidemia, non-alcoholic fatty liver disease. Globally, about 600 million people have been diagnosed as clinically obesity, which brought great social and financial burden, especially developed countries. Although substantial efforts have been made for development of anti-obesity drugs in recent decades, however, only two drugs are currently approved in the US for the long-term treatment of obesity: orlistat and sibutramine. Thus, new anti-obesity drugs are greatly needed. Lingzhi, as one of the most highly regarded medicinal fungi in the world, has a long medicinal history in China. Lingzhi was first recorded in Shennong's Herbal Classic and was listed as the highest grade. The ancients believed that Lingzhi was effective for improving health and extending lifespan after long-term consumption. G. lucidum is the most widely used species in Ganoderma, and it was documented in Chinese Pharmacopoeia (2020 edition). Pharmacological research revealed that G. lucidum has unique advantages in the treatment of obesity, because it can markedly reduce body weight, recover serum lipid homeostasis, inhibit hepatic lipid accumulation and steatosis, and improve insulin sensitivity and glucose metabolism in high-fat diet-induced (HFD) obese mice. Meanwhile, research has found that lanostane triterpenoids, as one of the major active constitutes of Ganoderma, play a key role in its anti-obesity effect. G. applanatum is another common species of Ganoderma. Although the structures of triterpenoids isolated from G. applanatum are different from that of G. lucidum, we found that the triterpenoid-enriched fraction (GAE) of G. applanatum can significantly inhibit 3T3-L1 preadipocyte differentiation and adipogenesis at 25 μg/ml. Therefore, this paper selected G. lucidum and G. applanatum as the research objects, in order to search lanostane triterpenoids with potential application in the treatment of obesity. Using modern separation technology, 105 triterpenoids were isolated from G. lucidum and G. applanatum, including 61 new compounds. These triterpenoids are characterized by various substituents, highly oxidized state and high unsaturation. Ganoderma triterpenoids can be divided into four subtypes according to the substituents at C-8, namely 8-dehydro-lanostane (1?69), 7,8-epoxy-lanostane (70?97), 8-hydroxyl-lanostane (98?103) and 8-oxo-lanostane triterpenoids (104?105). Among them, 8-oxo-lanostane triterpenoids were first reported in Ganoderma. Furthermore, we also found some unusual nortriterpenoid skeletons, including 20,21,22,23,24,25,26,27-octanorlanostane triterpenoid (1) and 2-norlanostane triterpenoid (70).
