摘要 | Bupleurum dracaenoides Huan C. Wang et al., published in 2013, is a species of family Apiaceae. Its typical morphological characteristic is the woody stem up to 1.5m compared to other species of Bupleurum. Its leaves grow in cluster at the top of stem, which are similar to those of Dracaena. It is the only woody species of the genus Bupleurum found in the Asian, and endemic to the dry and hot valleys of the Jinsha River. The distribution of B. dracaenoides is quite narrow, with only a few of populations found in Huidong County of Sichuan Province and Pudu River of Yunnan Province. It’s a critically endangered species, because the total number of individuals is very limited and its wild habitat is threatened. It is urgent to assess the genetic diversity so as to protect its wild resources efficiently. In this study, 153 samples from eight populations were studied to analysis the population genetics of B. dracaenoides with seven chloroplast DNA (cpDNA) markers and two nuclear DNA (nDNA) markers. Our aims were to make a reasonable evaluation for genetic diversity of B. dracaenoides and provide the theoretical strategies for the conservation of B. dracaenoides. Meanwhile, we preliminary explored the tissue culture technology of B. dracaenoides in this work. With the short stems with axillary bud selected as explants, the tissue culture technology for B. dracaenoides was preliminary explored through screening of disinfection methods, media types and different plant growth regulators. So that, we can provide a technical support for the artificial propagation of B. dracaenoides. The main results are as follows: 1. Evaluation of the genetic diversity of B. dracaenoides Seven cpDNA fragments were selected and evaluated as markers with high DNA variation level through sequencing, assembly and comparison six complete chloroplast genomes of B. dracaenoides. At the same time, two nDNA sequences(ITS and PZ8)were also selected as markers in this study because of their high variation in B. dracaenoides. Using the DNA data of the above markers, we analyzed the genetic diversity, genetic structure and demographic history of eight populations of B. dracaenoides. The results of cpDNA data analysis showed that there were high genetic diversity and high level of genetic differentiation among the populations of B. dracaenoides, and the genetic variation mainly existed among populations. But the genetic diversity was zero within a few populations. The results of nDNA data analysis showed that there was a relatively low level in genetic diversity and genetic differentiation among the populations, and the genetic variation mainly existed within populations. Haplotypes with closer genetic distances of cpDNA and ITS were clearly clustered into the geographically closer populations, which indicated that B. dracaenoides had a significant phylogeographic structure.
