绒毛诃子的化学成分和生物活性研究; Studies on Chemical Constituents of Terminalia chebula var. tomentella and Their Biological Activities

摘要Terminalia is the second largest genus in the family Combretaceae, mainly distributed in southern Asia and the Himalayas. There are about 250 species in the world, and nearly 40 species are used as medicines. The dried ripe fruits of Terminalia plants T. chebula Retz and T. chebula Retz. Var. tomentella Kurt are used in the Chinese Pharmacopoeia as Fructus Chebulae (FC). FC has extremely high folk application value, and is known as the "King of Tibetan Medicine". It can be combined with the fruit of T. bellirica Roxb and Phyllanthus emblica Linn to form Triphala, which has been the most frequently used prescription in Ayurveda and Tibetan medicine to play the role of treatment and preventive health care. In the past two decades, phytochemical and biological studies on plants of the Terminalia genus have been achieved great progress. At present, more than 120 compounds have been identified from T. chebula, mainly hydrolyzed tannins and triterpenoids, such as representative compounds chebulagic acid, chebulinic acid, punicalagin, arjungenin, arjunolic acid, terminolic acid, and maslinic acid. Activity studies have shown that it has many biological activities such as cardiovascular protection, antibacterial, antitumor, anti-inflammatory, and hypoglycemic. However, there are few reports on the pharmacological activity and chemical substance basis of T. chebula var. Tomentella. In order to clarify the similarities and differences between this variety and its original variety, further dig and exploit the resources of Terminalia, provide a chemical basis and scientific guidance for its ethnic medicines purposes, the chemical constituents and the pharmacological activities of T. chebula var. tomentella were studied in this paper. The analytical method of polyphenols in the fruits of T. chebula var. tomentella was established by HPLC, and the main chromatographic peaks were identified, including 7 hydrolyzed tannins (10, 11, 13-15, 17, 19), 3 simple phenols (8, 9, 20), and an oleanolane pentacyclic triterpene (25). Using a variety of chromatographic methods, modern spectroscopy techniques, and calculations of ECD and X-ray single crystal diffraction, a total of 44 compounds were separated and identified, including 11 hydrolyzed tannins (6, 10-19), 6 Simple phenols (8, 9, 20-23), 7 oleanolane pentacyclic triterpenes (7, 24-29), 16 lignans (1-5, 30-40), 4 flavones (41-44), of which 1-7 are new compounds, and 24 compounds are isolated for the first time from the genus Terminalia. Studies have shown that T. chebula var. tomentella and T. chebula all contain such marked ingredients as hydrolyzed tannins punicalagin (10), chebulagic acid (11), and triterpenoids arjungenin (25), arjunolic acid (27). It provides a certain scientific basis for the use of the two species as Chebulae Fructus in the Chinese Pharmacopoeia.
