摘要 | As the process of social aging continues to intensify, the subsequent prevention and treatment of neurodegenerative diseases represented by Alzheimer’s disease has become a severe social and public health problem. Neurotrophic factors with neuroprotective effects are important potential drugs for the treatment of neurodegenerative diseases. The resinous heartwood of Aquilaria sinensis (Lour.) Spreng. (Thymeleaceae) is used as a traditional Chinese medicinal, named agarwood (chen-xiang in Chinese). Our preliminary screening found that agarotetrol, the main component of agarwood, can promote the differentiation of PC12 cells. To further clarify the active ingredients in agarwood, we studied on chemical composition and its PC12 differentiation-promoting activity. And it also shows that agarwood formation may be closely related to the endophytic fungi. To understand its formation mechanism and better guide production, we have isolated and identified and screened the endophytic fungi in the resin and health woods. Using a variety of chromatographic methods and structural analysis techniques, 22 compounds were isolated and identified from the ethanolic extract of chen-xiang. Compound types include 2-(2-phenethyl) chromones, sesquiterpenoids, phenylpropanoid glycosides, and others. Among them, nine compounds are new, including six 2- (2-phenethyl) chromones, namely (5S,6R,7R,8S,7'R)-7'-hydroxyagarotetrol (1), (5S,6R,7R,8S,7′S)-7′-hydroxyagarotetrol (2), aquilarisinensin A (3), aquilarisinensin B (4), aquilarisinenone A (5), and aquilarisinenone B (6), along with three sesquiterpenoids, namely (4S,5S,7S,8S,10S,13R)-7,8,13-trihydroxyrotunda-1,11-dien-3-one (7), (4S,5S,7S,8S,10S,13S)-7,8,13-trihydroxyrotunda-1,11-dien-3-one (8), and (4R,5S,7S,8S,10S,13S)-7,8,13-trihydroxyrotunda-1,11-dien-3-one (9). All of the isolated compounds were tested for NGF-induced differentiation of PC12 cells. The result showed three 2-(2-phenethyl) chromone dimers (5, 17, and 18) had better activity. At a concentration of 10 μM, the differentiation rates of PC12 cells by compounds 5, 17, and 18 were 11.09%, 12.04%, and 14.18%, respectively, as compared with the positive control NGF with the differentiation rate of 19.32% at a concentration of 50 ng/mL. The activities of 5,6,7,8-tetrahydro-2-(2-phenethyl)chromone dimers are better than monomers. To further guide the production and find the strains that can promote the production of chen-xiang, we carried out the isolation of endophytic fungi on the fresh white wood resin and healthy parts. A total of 39 strains of endophytic fungi were isolated, of which 36 strains were isolated from the health wood, and only 3 strains were isolated from the healthy woody wood block. These endophytic fungi belong to 12 genera, of which the dominant dominant genera are Fusarium (30.77%), Nigrospora (20.51%), Nodulisporium (11.11%), and Hypoxylon (11.11%).
