药用狗牙花中单萜吲哚生物碱的研究; Studies on Monoterpene I ndole A lkaloid s from Tabernaemontana bovina

摘要Monoterpene Indole Alkaloids (MIAs) possess multiple skeletons endowing their diverse biological activities. To date, vinorelbine, ellipticine and camptothecin have been obtained the abundant achieveme nts in the clinic which may be the pharmaceutical value for researching MIAs . Based on it, MIAs have been hotspot in different academic sector and researcher. Our group have been endeavored to explore MIAs and provided with experiences about isolation and structure elucidation of them in last decade. These were the main reasons that we carried out the systematic investigations on MIAs from Tabernaemontana bovina and 262 non repeating MIAs have been isolated, including 132 unreported MIAs This thesis inclu ded 7 chapter s and clarified the five skeleton types of MIAs and their partial biological activities. T he taberbovine that originate d from the condensation of tryptophan with secologanin in a 2:1 ratio was isolated in first time from the Tabernaemontana pl ants. The changeable functional in the indole A ring of aspidospermine were improved and supplied. Moreover, there were unreported skeleton rearrangement in the indole A ring and rarely polycyclic bird caged alkaloid with an oxepane bridge ring simultaneou sly were revealed . In the type of iboga, we found two pair of enantiomerics that have been firstly reported about MIAs in the same plant so far. With respect to bisindole (dimeric MIAs), we also continuously obtained the furan aspidospermine that possessed potential tumor activities and two aspidospermine units were unprecedentedly assembled though C C single bond Then, taberbovtines D E and taberbovtines F G were isolated as brand new bisindole that contained units of vincamine aspidospermine and pleiocar pamine aspidospermine, respectively. Furthermore, tabernaemontines N V p ossessed a xial chirality by connect ion of single bond at C9 C9′ which w as further unique. Additionally, we tried to establish furan dimeric by organic synthesis The absolute configura tion of above new MIAs were determined by single crystal X raycrystallographic experiment and quantum computation (ECD and NMR). And then about MIAs’ bioactivities, we surprisingly found tabernaemontines G showed good inhibition of microglial activation t hough blocking P 38 MAPK activation. Meanwhile, voaphylline could promote the glucose uptake in L6 myoblasts by the hypoglycemic experiment and others bioactivit ies are working. A series of novel MIAs were excavate d from Tabernaemontana bovina and their se ries of s tructures were diversely characterized , which demonstrated glamourous of natural products again . The discovery of new bioactivit ies alkaloids aimed at nervous system and metabolic diseases exhibited the functional attractive s. Therefore, the thesi s was like to throw out a minnow to catch a whale and we willexpect to research enormous potential of MIAs with more peers
