几种兜兰属植物的种子无菌萌发和组织培养; Studies on asymbiotic germination and tissue culture of several Paphiopedilum species

摘要Members of the genus Paphiopedilum are world famous ornamental orchids. At present, over-collection and loss of natural habitat have drastically reduced wild populations of Paphiopedilum species. All members within Paphiopedilum have been listed in Appendix I of the Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES), and are protected at the highest level. The breakthrough of seedling propagation technology is of great significance to the germplasm utilization and protection. Asymbiotic seed germination and tissue culture are considered as effective means for orchid propagation. However, tissue culture of Paphiopedilum plant is very difficult to overcome. Although the tissue culture technology of a few Paphiopedilum species has made some progress at present, it is still far from commercial application. Seed asymbiotic germination has been successful on many Paphiopedilum species, but there are great differences among species. Seed asymbiotic germination techniques of some species have still not been overcome, such as P. tigrinum. This is a rare species with high commercial value, but the extremely low germination rate of seeds limits its large-scale production and reintroduction to native habitats. In the present study, we used P. appletoniantum, which is easier to germinate, as a control, and studied the effects of basic media, activated charcoal, hormones, organic matters, and light on the seed asymbiotic germination and seedling growth of P. tigrinum. The purpose was to find out the key factors affecting the seed asymbiotic germination and seedling growth of P. tigrinum, and to establish seed asymbiotic germination technique of P. tigrinum with high germination rate and plantlet regeneration rate. Meanwhile, we used the stem segment of Paphiopedilum Complex Type as explant to study the effects of explant size, plant growth regulators and other factors on its plantlet regeneration, and attempted to make advances in tissue culture techniques of Paphiopedilum. The results were shown as follows. 1. There was significant difference in the requirement for the components of the culture medium between Paphiopedilum tigrinum and P. appletoniantum, and among developmental stages. At the seed germination stage, the type of basic medium had an effect on the seed germination of P. tigrinum, but had no effect on P. appletoniantum. The seeds of P. tigrinum showed the highest germination rate on modified Harvais (cHa) medium. The addition of 0.1 g/L activated charcoal or 0.5 mg/L kinetin (KT) promoted seed germination to a certain extent. The seed gemination rate of P. appletoniantum on cHa, half-strength Murashige and Skoog (1/2 MS) and 1/4 MS medium was not significantly different, but was promoted by the addition of 0.5~1.0 g/L activated charcoal or 0.5~1.0 mg/L α-naphthalene acetic acid (NAA).
