摘要 | This thesis consists of five chapters. High performance liquid chromatography mass spectrometry based metabolomics is widely used in modern pharmaceutical analysis The first chapter described the application of HPLC-MS in the chemical composition analysis and quality control of traditional Chinese medicine; The second chapter elaborated the application of metabolomics in the quality marker of Citrus aurantium; The third chapter illustrated the application of metabolomics in quality marker of Gardenia jasminoides; The fourth chapter elucidated the application of comparative metabolomics in the metabolism and toxicity of geniposide and genipin; The fivth chapter elucidated the application of metabolomics in the regorfenib metabolism. The chemical composition of Chinese herbal medicine is various and complex, the quality of Chinese herbal medicine is unstable, and there is no effective quality control. High performance liquid mass spectrometry has the characteristics of high sensitivity and high separation, which is suitable for the analysis and quality control of complex Traditional Chinese medicine. In this chapter, the application of HPLC-MS in chemical composition analysis and quality control of traditional Chinese medicine is reviewed, which provides reference for the research of chemical composition analysis and quality control of traditional Chinese medicine. Citrus aurantium is a kind of traditional Chinese medicine for treating indigestion. Citrus aurantium produced in Jiangxi Province is genuine medicinal materials. The quality markers of Citrus aurantium was screened in the present study. In this study, a novel solid phase extraction- mass loss- molecular network strategy was established, and 57 chemical components of Citrus aurantium were identified by two-stage mass spectrometry. The different chemical constituents of Citrus aurantium in Jiangxi Province and other two non-authentic regions were screened by comparative metabonomics. The absorbed components were screened by serum pharmacochemistry under physiological conditions and pathological conditions of indigestion. The common constituents between different constituents and absorbed components were selected, and further tested their inhibition on peripheral acetylcholinesterase. The results showed that neohesperidin, naringin and meranzin could inhibit the activity of acetylcholinesterase, and molecular docking simulated the interaction between the above three compounds and acetylcholinesterase. The neohesperidin, naringin and meranzin were identified as potential quality markers of Citrus aurantium. Gardenia jasminoides is a kind of traditional Chinese medicine for treatment of jaundice. Gardenia jasminoides produced in Jiangxi Province is genuine medicinal materials. The quality markers of Gardenia jasminoides from Jiangxi Province was screened in this study.
