摘要 | Paris polyphylla Smith var. yunnanensis (France). Hand-Mazz, a perennial herb, belongs to the genus Paris (Melanthiaceae). And it has been officialized as one of the original plants of Rhizoma Paridis (Chonglou) by Chinese Pharmacopoeia (1977-2015 Editions). This herb has been widely used as a folk medicine possessing the effects of clearing away heat and detoxification, relieving swelling and pain, calming the liver in China since ancient times, and is one of the main active ingredients in many Chinese patent medicines. This genus was divided into Daiswa and Paris two subgenera. The original plants of the Chinese Pharmacopoeia belong to the subgenus Daiswa, wheras the subgenus Paris used as herb medicines in the folk. However, there are no reports of the material basis and bioactivities on the subgenus Paris. Therefore, aiming at protecting and utilizing reasonably the subgenus Paris, the phytochemical and bioactivities of Paris tengchongensis and Paris rugosa have been investigated based on a method combining the LC-MS/MS molecular networking strategy with the conventional means of phytochemical research. This dissertation consists of two chapters. The first chapter detailed the research of phytochemical and bioactivities of P. tengchongensis rhizomes. Firstly, we described the molecular network technique, which was used to recognize and mine the chemical components in P. tengchongesis. A MS/MS data of sample collected by UPLC-Q/TOF to generate a molecular network in GNPS (https://gnps.ucsd.edu). Then the visualization results were disposed with a software named Cytoscape. As a result, the chemical constituents of similarities and differences between P. tengchongesis and P. polyphylla. yunnanensis were illustrated. Secondly, the chemical constituents from the 80% EtOH extract of P. tengchongesis were investigated. A total of 25 compounds, including 23 steroidal saponins (4 new ones) were obtained and characterized based on the molecular network analysis by means of various methods like macroporous resin, silica gel, Rp 18 silica gel, Sephadex LH-20, semi-preparative HPLC, combined with NMR, MS and hydrolysis techniques. These isolates consist of 16 spirostane-type saponins (1–3, 5–17) and 7 furostane-type saponins (4, 18–23). The sugar chains are consisted of β-D-Glu, α-L-Rha, α-L-Ara, β-D-Xyl, β-D-Fuc and β-D-Api. Compound 1 is a new polyhydroxylated steroidal glycoside with a disaccharide chain attached to C-1 and C-24, while 2 and 3 have a new sugar chain: β-D-xylopyranosyl-(1→6)-[β-D-glucopyranosyl-(1→2)]-β-D-glucopyranoside. Compound 4 is a new furostane-type glycoside and is the protodioscin of gracillin. The new isolates 1–3 were evaluated for their cytotoxicities against two human cancer cell lines (HEL and MDA-MB-231), anti-inflammatory effects on LPS-stimulated NO production in RAW264.7 macrophages, anti-AChE, and antimicrobial activities. |
