摘要 | Bamboos (Poaceae: Bambusoideae) have a unique life history, they have long reproductive cycles from a few up to 120 years and low spikelet fertility, and their vegetative propagation mainly depends on the lateral growth of rhizomes. Rhizome is an important taxonomic character of bamboos, however, the factors that regulate the growth and development and the evolutionary mechanism of the rhizome types are still unknown. Starting from investigating the morphology of four rhizome types of bamboo, in this study we excavated and observed the morphological differences among different rhizome types, and observed the development characters of lateral and terminal buds of monopodium and amphipodium bamboo rhizomes using paraffin sectioning method. We also statistically analyzed the probability of bamboo "clumps" of these two rhizome types. In addition, due to the similarity between sympodial bamboos and the tillering growth of other plants in Poaceae, we selected 17 orthologous genes with obvious differences among 51 genes related to tillering, and compared the structural differences of these genes among different rhizome types, so as to preliminarily understand the genetic basis and development pattern of different rhizome types of bamboo. The mainly results and conclusions are as follows: 1. Observation results on different rhizome types of bamboo The lateral buds bear on the monopodium and amphipodium rhizomes can develop into new shoots or rhizomes, and the rhizomes can further develop into an interconnected network underground. Paraffin sections were performed on lateral and terminal buds of the rhizomes of Indocalamus tessellatus and Phyllostachys edulis. We found significant differences in structures between lateral buds which developed into bamboo shoots or rhizomes. Shoot buds have a dense "ring" like areas of cells in the pith, which will develop into nodes in the future, and the nodes’ number can be clearly seen. Although there are also dense areas of cells in rhizome buds and terminal buds, they look like "strips" in the longitudinal direction and the number of segments cannot be seen. This difference also proves that the growth and development patterns of bamboo shoots and rhizomes are different, that is, the growth of bamboo shoots is limited while the growth of rhizomes is infinite. 2.Statistics of clumps of monopodium and amphipodium bamboo rhizomes Statistics of clumps were analyzed among different rhizome types, Acidosasa, Brachystachyum, Hibanobambusa, Phyllostachys and Sinobambusa were sampled as representatives for monopodium bamboos, and Chimonobambusa, Indocalamus, Pleioblastus, Shibataea, Sasa and Sasaella were sampled as representatives of amphipodium bamboos. According to the statistical results, the probability of "clumps" in monopodium bamboos is much lower than that in amphipodium bamboos. Even with very small probability, the fact that lateral buds bear on culm-base of monopodium bamboos can form culms proved that bamboo shoot |
