旱生香茶菜中两株内生真菌的次生代谢产物研究; Studies on the secondary metabolites of two endophytic fungi from Isodon xerophilus | |

摘要 | The bioactive secondary metabolites of microorganism are always a significant leads source in drug discovery. Endophytic fungus, a unique type of microorganism possessing species, gene and metabolites diversity due to effects of hosts metabolites, have become a source of novel bioactive secondary metabolites. However, on account of the enormous endophytic fungi have great discrepancy between their species, how to screen out the “Talent fungus” which can produce structurally abundant and novel metabolites has raised universal concern of researchers. Meanwhile, some studies, which are associated with the efficient and convenient methods like “LC-MS/MS-GNPS”, have been applied into excavation of “Talent fungus”. Our laboratory has conducted in-depth investigation about screening strategy of “Talent fungus” in some plants of the genus Isodon previously. In this dissertation, Isodon xerophilus was selected as the host plant to explore its endophytic fungi by systematic screening strategy based on literature research, metabolites abundance and chemical space analysis. 806 strains have been isolated and purified from Isodon xerophilus. These strains were subsequently classified as 10 genus and 29 species by morphological comparison and ITS identification. Eventually, two strains named Pestalotiopsis sp. HS30 and Arthrinium sp. HS66 were selected for further chemical investigation with systematic screening. On the basis of chromatographic fractionation, spectroscopic analysis and signal crystal X-ray diffraction analysis, 55 compounds including 32 new ones which contained two alkaloid epimers (Pes1 and Pes2) featuring an unprecedent skeleton. In addition, both compounds Pes1 and Pes2 showed the latent tumor immunotherapy activity through markedly promoting the cell surface engagement of NKG2D ligands in HCT116 cells on the course of bioactivity evaluation. Therefore, the result of this dissertation further perfected the systematic screening strategy and confirmed its applicability and reliability of targeting “Talent fungus”, which would promote the development of investigation on secondary metabolites from the endophytic fungus which inhabited the Isodon species. |
