摘要 | Abnormal lipid metabolism refers to a physiological and pathological process in which lipid substances are abnormally synthesized, decomposed, digested, absorbed, and transported in the body, resulting in too much or too little lipid in various tissues, which affects the body's functions. With the rapid economic growth, the diet of people has changed greatly. Long-term intake of high cholesterol, high saturated fatty acids and high-calorie diets and lack of exercise lead to disorders of the lipid metabolism. Disturbance of lipid metabolism increase the risk of chronic diseases such as obesity, fatty liver, hyperlipidemia, and atherosclerosis. In this work, the pharmacological activity and molecular mechanism of two natural terpenoids are investigated for obesity and atherosclerosis caused by excessive lipid accumulation. As lifestyle changes, the population of obese and overweight continues to increase. Obesity is a high-risk factor that leads to various chronic diseases, including cardiovascular disease and hypertension, which has brought a serious medical burden to the society. At present, most of anti-obesity medications were withdrawn from the market and not widely used due to their inefficiency or undesirable side effects. Therefore, the development of therapeutic drugs with fine curative effect, minor adverse effects and high safety is the focus of current research. Eriocalyxin B, an ent-Kaurene diterpenoid extracted from a traditional Chinese herb Isodon eriocalyx, has been shown to possess multifunctional activities including anti-cancer and anti-inflammatory. However, the function and mechanism of the compound in adipocyte differentiation is still unknown. Therefore, we explored the pharmacological effects of Eriocalyxin B in adipogenesis, and the experimental results show that Eriocalyxin B blunted adipogenesis remarkably by inhibiting the accumulation of lipid droplets, triglycerides and the expressions of adipogenesis-related factors, including C/EBPβ, C/EBPα, PPARγ, and FABP4. Moreover, we showed that the inhibition might be the consequence of cell cycle being arrested at the G2/M phase during the mitotic clonal expansion of adipocyte differentiation, most likely by suppressing mRNAs and proteins of CDK1, CDK2, Cyclin A and Cyclin B1. In conclusion, we conclude that Eriocalyxin B is capable of inhibiting adipocyte differentiation at the early stage through downregulating the proteins involved in cell cycle progression. Borapetoside E, a clerodane diterpenoids extracted from the medicinal plant Tinospora crispa. Our previous research reported the clear structure of borapetoside E, and further reported that borapetoside E could significantly improve the hyperglycemia, insulin resistance, hyperlipidemia, fatty liver, obesity, and energy metabolism in type 2 diabetic mice.
