摘要 | Abstract High-Speed Countercurrent Chromatography (HSCCC) is an advanced liquid-liquid partition chromatography separation technology developed in the 1980s, and it is prevalent in natural product research over the recent years. During the past three decades, our lab has undertaken phytochemical investigations on Schisandraceae plants by employing a variety of separation approaches. However, all these conventional techniques had their own limitations. For instances, irreversible adsorption often occurs in silica gel column chromatography; limited choices of eluting solvents were available in reverse-phase silica gel column chromatography; and the separating effect of gel column chromatography heavily depends on the differences in molecular weights of compounds to be separated. Thus, to overcome these problems, as well as to further promote the understanding and application of the chemical constituents from Schisandraceae species, it is urgent to find new separation techniques. The HSCCC came into our sight for it can run without fixed carriers and peak trailing, and the crude extract can be separated, purified, and prepared on a large scale simultaneously with HSCCC. Thus, this project aims to obtain abundant and diverse compounds from Schisandraceae species, with an emphasis on the employment of HSCCC approach. This thesis has described the rapid separation of active components (schisandrins A–C, schizandrol) from an important medicinal plant, Schisandra chinensis through HSCCC, as well as the utilization of HSCCC technology in phytochemical investigation of S. propinqua var. propinqua. Chapter one: The application of HSCCC in Separating and Enriching Bioactive Lignans from S. chinensis (Turcz.) Baill. S. chinensis (Schisandraceae) is a deciduous woody vine mainly growing in valleys, streams and hillsides at 1200~1700 m altitude, and it is widely distributed in northern China, North Korean, Japan and Russian far East areas. In traditional Chinese medicine, the dried ripe fruit of S. chinensis is a famous medicinal material (“wuweizi”) mainly used as tonic and sedative agents. Dibenzocyclooctadiene lignans are among the major constituents in S. chinensis, and they usually possess favorable bioactivities. Noteworthily, the content of schizandrol, a dibenzocyclooctadiene lignan, is a standard for quality control of S. chinensis in Chinese Pharmacopoeia. To enrich the bioactive lignans in S. chinensis is of great significance, not only for drug research, but also for the exploitation and utilization of Schisandraceae resources in China. Accordingly, an effective method was established to rapidly separate and purify diagnostic lignans (schizandrins A–C, and schizandrol) from S. chinensis acquired from Yanji City by using HSCCC, Jilin Province.
