橡胶树重要种质资源精准评价与多倍体种质创制; ACCURATE EVALUATION OF IMPORTANT GERMPLASM RESOURCES OF RUBBER TREES AND CREATION OF POLYPLOID GERMPLASM

摘要The botany character and latex physiological properties of important wild and cultivated germplasm of Hevea brasiliensis (rubber tree) were carried out by precise evaluation, we found that the botany character and latex physiological properties of wild and cultivated germplasm of rubber tree are very rich in variation. For most traits, variationamplitudeofwild germplasmisgreaterthanthecultivated germplasm, there is still relatively rich genetic information in wild germplasm, it could be used in thebreeding. Fresh seeds and germinating seeds of rubber tree varieties GT1, RRIM600 and IAN873 were treated with colchicine, A total of 20430 mutagenic M1-generation seedlings were obtained. 38 M1-generation seedlings containing tetraploid cells were detected, but all tetraploid seedlings were chimeras. Eleven triploid germplasm were detected from M1 generation seedlings of GT1, and the triploid yield was 0.31%, it providinganewwayforsubsequentpolyploidbreedingofrubbertrees. The method was established to determine thegenomic C value of rubber trees. It was found that 11.32% (6 germplasms) of the germplasms which have good performance advantages in rubber plantations in Yuannan province were triploid, much higher than the rate of plant polyploidy in the natural enviroment. This indicates thatthetriploidgermplasmofrubbertreehasagoodperformance advantage in the high altitude rubber plantation area of Yunnan province, which makes it be screenedoutintheobservationandevaluationformanyyears. The genome of rubber tree GT1 was sequenced by SMRT and Hi-C technology, and about 1.47 Gb of the assembled genome sequence was mounted on 18 pseudochromosomes. The chloroplast genomes of Hevea camargoana, Hevea bethamiana and 12 wild and cultivated Hevea brasiliensis germplasms was sequenced, the results showed that the chloroplast genome size of rubber tree was 161,168 bp-161,254 bp, all containing 112 genes, the DNA coding region of the chloroplast genome evolved more slowly than the non-coding region. Evolutionary analysis showed that there was a close genetic relationship between wild related species,wildgermplasmandcultivatedgermplasmofrubbertree.
