大苞鞘石斛化学成分研究及石斛化学分析; Study on Chemical Components of Dendrobium wardianum and Chemical Analysis of Dendrobium

摘要This thesis covers four chapters. The research of the chemical composition of Dendrobium was carried out aim to further understand the similarities and differences among different species of Dendrobium. We can understand the distribution characteristics and biosynthetic parts of main secondary metabolites, and provide useful scientific basis for the rational traditional Chinese medicine of Dendrobium. The first chapter is about the phytochemical separation, purification, structure analysis and cytotoxic activity of the Dendrobium wardianum Warner. Meanwhile the fat-soluble components of D. wardianum of roots, stems and leaves were analyzed. Total eighteen compounds were isolated and identified. A variety of chromatographic separation techniques and semi-preparative liquid chromatography were used to separate and purify compounds. And the structural types of compounds involves phenylpropanoids, phenanthrenes, sesquiterpenes, flavonoids, bibenzyls and steroids, they were identified by NMR , MS, TLC et. Among them, compounds 3, 5 and 6 were first isolated from Dendrobium, and compounds 10, 11, 14 and 15 were first isolated from this species. Then some phenolic compounds 3, 6, 8, 9, 11, 14 and 15 were screened for cytotoxic activity using MTS method on five tumor cells. The results showed that compound 6 and 8 had better cytotoxic activity, IC50 value of 0.908~8.72 μM. The second chapter is to analyze the different chemical constituents of 16 species of Dendrobium by HPLC-UV. HPLC datas were imported into the similarity evaluation system of traditional Chinese medicine chromatographic fingerprint (version 2012.130723). The results showed significantly distinguish in those species, which was in accordance with the differences of the chemical constituents obtained from recent literature. The difference between the twelve species of Sect. Dendrobium was relatively large. But the difference between the two species of Sect. Formosae was relatively small and the two species of Sect. Chrysotoxae was insignificant. The results of this analysis provided a basis for further reasonable application of Dendrobium and the reclassification of Dendrobium. In chapter three, the progress on distributions and biological activities of alkaloids and sesquiterpenes in Dendrobium was reviewed. And the phytochemical taxonomy of this genus was further discussed, providing a basis for the potential application of Dendrobium. In chapter four, we summarized this paper.
