摘要 | Ginseng (Panax ginseng C.A. Mey.) is a valuable Chinese herbal medicine with high economic value. Ginseng is mainly propagated by seeds, however the embryo is morphological hypoplasia when ginseng seeds abscission occurred, seeds having the characteristic of dormancy, and the dormancy period up to 9-22 months, which is very unfavorable to the production of ginseng. The embryo is an important part of seed, and after-ripening of the embryo is very important for ginseng seeds to break dormancy. Although the variable temperature can promote the after-ripening and germination of ginseng seeds, the molecular mechanism of the after-ripening process is still unclear. Therefore, in this study, seeds of Panax ginseng C.A.Meyer cv. Silvatica were stratified in stages at 16℃ and 4℃ to explore the impact of temperature on seed germination, and morphological changes of seeds during after-ripening. Transcriptome analysis of embryos at different after-ripening stages was implemented to explore the gene expression regulation, to reveal the molecular mechanism of embryo after-ripening. Our study will provide reference basis for research dormancy-release and long-term preservation of ginseng seeds. The main results are as follows: 1. Our results of temperature stratification showed that the 16℃ treating could promote morphological after-ripening (the morphological development process from heart-shaped to mature embryo) of embryos and seeds cracking for ginseng, and the 4℃ treating can promote the physiological after-ripening and germination of ginseng seed. Ginseng seeds were stratified at 16℃, which seeds can be split in about 90 days and mature morphological in about 180 days. After that, it took about 90 days for the seeds to be stratified at 4℃ to complete physiological ripening and germination. It took about 270 days in total for ginseng seeds from stratification to germination. Our study result has important meaning to direct ginseng seedling. 2. We used the Illumina HiSeq X-ten platform to sequence the transcriptome of ginseng embryos at five different after-ripening stages: heart-shaped embryo (0 days), torpedo-shaped embryo (45 days), cotyledon embryo (90 days), mature embryo (180 days) and bud embryo (270 days), which generated 412604390 raw reads in total, and 404850531 clean reads were obtained after quality control. When clean reads of each sample were mapped to ginseng genome, the mapping rate reached more than 94%. 3. When compared to heart-shaped embryos, transcriptome comparing analysis revealed 2359, 3048, 6180 and 12067 differentially expressed genes (DEGs) in torpedo-shaped embryos, cotyledon embryos, mature embryos and bud embryos respectively. Gene Ontology(GO) enrichment analysis showed that the DEGs were all significantly enriched into pathway related to catalytic activity, binding, cell, cellular process, metabolic process and developmental process. 4.
