摘要Natural products and their derivatives play important roles in prevention and treatment various human dieases. The isolation of morphine from Papaver somniferum is a landmark of the beginning of searching for active ingredients from natural products and also the initiation of modern natural medicinal chemistry. Natural products from plants have made outstanding contribution to the development of natural medicinal chemistry. There are some drugs, such as artemisinin and taxol, could be directly acquired from plants, while, most other drugs could only be indirectly obtained. For example, the anti-cancer drugs irinotecan and topotecan were successfully developed through structural modification of camptothecin, and vinorelbine was designed and developed based on vinblastine and vincristine. Thus, the purpose of this dissertation was to excavate active natural products of plants by multiple isolation and identification techniques and improved the bioactivity of natural compounds through chemical modifications. This dissertation consists of four chapters. Chapter 1 described the systematic investigation on the secondary metabolites of the aerial parts and roots of Phlomoides betonicodes (Lamiaceae) and their biological activities. Chapter 2 illustrated the study on secondary metabolites of the leaves of Plukenetia volubilis (Euphorbiaceae). Chapter 3 dealt with the structural modification of leucosceptriods A, G, N and immunosuppressive assay. Chapter 4 reviewed the research progress on diterpenoid glycosides in the last three decades, invloving their sources, structure types, biological activities and biosyntheses. Through multiple chromatographic techniques and modern spectroscopic methods, a total of 222 compounds of diversified structure types, including triterpenoids, diterpenoids and their glycosides, iridoid glycosides, sesquiterpenoids, flavonoids, coumarins and alkaloids, were isolated and identified from P. betonicodes and P. volubilis. Among them, 46 compounds were new, including nortriterpenoids, diterpenoids and their glycosides, iridoid glycosides, sesquiterpenoids and coumarins. Through chemical derivatization of leucosceptriods A, G, N, 54 modified sesterterpenoids with new structures were obtained. Their in vitro inhibitory effects of some synthesized compounds on anti-CD3/CD28 monoclonal antibodies (mAbs) induced cytokine IFN-γ production of mice primary T cells were tested.
