摘要Craigia yunnanensis W W Smith & W E Evans in Flora of China is a deciduous tree of genus Craigia WW Smith & W E Evans in family Tiliaceae (Flora of China) or Malvaceae (APG IV classification system). As a threatened plant species, C. yunnanensis was listed as the grade-II protection in China’s national key protected wild plants (First Group) in 1999, and it has also been targeted as PSESP (Plant Species with Extremely Small Populations) for urgent rescuing conservation in both lists of China and Yunann province. C. yunnanensis is not only important for scientific research and conservation, but it’s also a valuable timber tree, and an excellent plant for degraded ecosystem restoration in the karst regions. The study on conservation biology of C.yunnanensis has being carried out since 2005. Based on the current progress on conservation works of C.yunnanensis, this study has developed SSR primers used to explore its genetic diversity. Based on the analysis of the parents of the populations with different levels of human disturbance to reveal the pollen flow and mating system, as well as pollen dispersal distance. In addition, the relationship between the mating system and seedling growth of offspring of C. yunnanensis was explored. The main findings are as follows: 1. Using MiSeq sequencing technology, 14 pairs of SSR primers with polymorphism and specificity were developed. By using the 14 pairs of primers, polymorphisms were detected in 5 populations from Wenshan and Dehong regions. 159 alleles were detected in 30 individuals from the populations, and the number of alleles (Na) at each locus was 5–18, with an average of 11; the observed heterozygosity (Ho) was 0.400–0.967, average 0.730; expected heterozygosity (He) was 0.671–0.898, average 0.805. Hardy–Weinberg equilibrium test showed 8 loci deviation. Thus, the developed primers can be used to study the genetic diversity, genetic structure and mating system of C. yunnanensis. 2. The 221 individuals from 19 populations of C. yunnanensis were sampled for analyzing the genetic diversity by using 11 pairs primers, and a total of 73 alleles were detected, of which 52 were effective alleles; the observed heterozygosity was between 0.182–0.909, and the expected heterozygosity was 0.091–0.715. Xiajinchang (XJC) population has the highest genetic diversity (Na = 10.000, Ho = 0.749, He = 0.794, I = 1.859, PPL = 100.00%).The population percentage of polymorphic sites was 100% in populations of Xiajinchang (XJC), Fadou (FD), Donggan (DG), Pingba (PB), Jiangdong (JD) and Lushui (LS). The Mantel test performed a correlation test on genetic distance and geographic distance, and significant correlation (R2 = 0.31, P = 0.01). UPGMA analysis showed that 19 populations could be roughly divided into 5 groups in GD0.6 as the threshold.
