肾茶的化学成分及生物活性研究; The chemical constituents and bioactivities of Clerodendranthus spicatus

摘要The whole plant of Clerodendranthus spicatus (Thunb.) C. Y. Wu (Labiatae) is one of popular tea in south of China, named as “kidney tea” and used for treatment of renal inflammation, bladder inflammation, gout, arthritis and so on. Some previous studies reported anti-inflammatory effect of the extract in vivo. In present investigation, ethanol extract, two fractions (ethyl acetate and water fractions from the ethanol extract) and subfractions (from the ethyl acetate fraction) were prepared and evaluated for their anti-inflammatory effect on xylene-induced mice ear edema model. Then, the bioassay-guided isolation of bioactive subfractions led to 25 compounds were obtained. Among them, compounds 2, 3, 5, 6, 7, 9–11, 13, 16, 17, 20, and 21 inhibited the productions of pro-inflammation factors TNF-α, IL-1β, and IL-8 in lipopolysaccharide (LPS) -induced renal epithelia (HK-2) cells, respectively (P < 0.01/0.05). Further anti-inflammation evaluation in vivo indicated that the major bioactive compounds 1, 2, 5–7, 17, 21 and 22 from C. spicatus were even better than aspirin. Meanwhile, the results suggested that highly oxygenated diterpenoids and polymethoxy flavonoids may be the material basis of their anti-inflammatory activity, and the importance of nonpolar substituent for its anti-inflammatory effect. Meantime, the study was also investigated the effect of ethanol extract and subfractions from C. spicatus to relieve gout by assessing the anti-hyperuricemic, anti-inflammatory, and analgesic activities. The serum uric acid (UA) level was examined in potassium oxonate (OXO)-treated mice and the effect of sodium urate crystal on the excretion of uric acid in HK-2 cells was determined, which evaluated the activity of anti-hyperuricemia. The effect of ameliorated swelling was investigated in monosodium urate (MSU) crystal -induced arthritis model, and the effect of analgesia was evaluated in acetic acid-induced writhing test. The results indicated ethyl acetate fraction (EAF) with bioactive of anti-inflammation, analgesia and anti-hyperuricemia in vivo, After then, the isolation of EAF led to 37 compounds were obtained (including 25 compounds from Fr.1 and Fr.2. Among of compounds 26 and 27 were new compounds. Further pharmacological investigation of pure compounds indicated that compounds 3, 4, 5 and 32 significantly promoted the excretion of uric acid in vitro. And compounds 1, 2, 4, 6, 8, 16, 21, 25, and 34 showed the remarkable anti-inflammation activity. Among of, compounds 1, 2, 4, 5, and 8 showed the significant analgesia activity (P < 0.01/0.05). The results indicated that C. spicatus relieve gout by regulating hyperuricemia, gouty arthritis and pain.
