摘要Serine carboxypeptidase-like protein (SCPL) is proteases belonging to a/β hydrolase family. Up to now, SCPL has been proved to be related to seed germination, protein transport and stress response from the physiological and cellular levels, but the related molecular mechanism is still poorly understood. In here, we identified and characterized the T-DNA mutant of serine carboxypeptidase-like 41 protein (SCPL41), named bis4 (1-butanol insensitive 4). Then we choosed Columbia wild-type (Col), SCPL41 overexpression lines (SOE-5 and SOE-7) and the mutant lines (bis4 and bis44), using transcriptome, proteomics and lipomics methods to explore the functions of the gene. The results showed that SCPL41 gene positively responded to ABA, and improved the drought resistance of plants and also participated in the regulation of Arabidopsis membrane Metabolism. The main conclusions are as follows: First, SCPL41 gene positively responses to exogenous ABA. Under the exogenous ABA treatment, after SCPL41 was be deleted, the SCPL41 deletion plants, bis4 and bis44, were insensitive to ABA, and their germination rate of seeds were significantly higher than that of wild-type plants. On the contrary, SCPL41-overexpression plants, SOE-5 and SOE-7, were hypersensitive to ABA, and their seeds germination rate was significantly lower than that of wild-type plants. The main root length of SOE-5 and SOE-7 plants was significantly lower than that of wild type plants. These results suggest that SCPL41 may positively response to exogenous ABA. Second, SCPL41 can enhance drought tolerance in Arabidopsis thaliana. Compared with wild-type Arabidopsis thaliana, bis4 and bis44 were insensitive to ABA inhibiting stomatal opening and promoting stomatal closure, while SOE-5 and SOE-7 were supersensitive to ABA inhibiting stomatal opening and promoting stomatal closure. The wild type, SCPL41 deletion lines (bis4 and bis44) and SCPL41 overexpression lines (SOE-5 and SOE-7) were naturally dehydrated at room temperature, and the water loss rate of bis4 and bis44 was significantly higher than that of wild type. The water loss rate of SCPL41 over-expression line (SOE-5 and SOE-7) was significantly lower than that of wild type. Under drought treatment, the bis4 and bis44 plants were more sensitive to water, and their leaves wilted. After 14 days of drought, all the plants died, but they did not survive after re-watering. The seedlings of SOE-5 and SOE-7 plants were insensitive to drought. After 14 days of drought treatment, the survival rate of plants reached 90%. The phenotype of wild-type plants was between the deletion and over-expression lines, and the survival rate was about 43.7%. Third, SCPL41 enhanced drought resistance of Arabidopsis thaliana through ABA-mediated signal transduction pathway. Fourth, SCPL41 negatively regulated membrane lipid metabolism in Arabidopsis thaliana.
